摘  要:以黑松无菌苗带子叶顶芽为外植体,从丛生芽的诱导、伸长、增殖及不定根形成等方面进一步优化直接器官发生和植株再生体系。结果表明:丛生芽诱导最佳组合为GD+3 mg/L 6-BA+0.3 mg/L NAA+25 g/L蔗糖,丛生芽诱导率为95.7%,平均每外植体芽数为3.1;在DCR+0.5 mg/L 6-BA+0.2 mg/L NAA+20 g/L蔗糖的培养基上丛生芽伸长明显,添加0.5 g/L活性炭对丛生芽伸长也有明显的促进作用;在GD、1/2 GD、DCR 3种基本培养基中,DCR培养基较适合黑松增殖培养;根伸长阶段用珍珠岩培养基,提高了再生植株的根系质量和移栽成活率;基因型对移栽成活率有一定的影响,30-B 1个月移栽成活率最高达80%。Using the aseptic no rooted plantlets of Pinus thunbergii as explants,plantlet regeneration via direct organogenesis was optimized from axillary buds induction,shoot elongation,proliferation and root formation.The optimal combination of axillary buds induction was GD medium supplemented with 3 mg/L 6-BA,0.3 mg/L NAA,25 g/L sucrose.The axillary buds induction rate was 95.7%,and the average buds number was 3.1 per explant.Shoots were elongated obviously on DCR medium containing 0.5 mg/L 6-BA,0.2 mg/L NAA,20 g/L sucrose or 0.5 g/L activated charcoal.Among the three basal media(GD,1/2GD,DCR),DCR medium was the best to the proliferation of axillary buds.Perlite medium could improve the root quality and the survival rate of plant regeneration in root elongation stage.Genotype had some effects on transplant survival rate,the transplant survival rate of 30-B was up to 80%.
