摘 要:目的:探讨人β-防御素2加强乙肝疫苗对小鼠免疫应答的作用。方法:采用乙肝疫苗2μg/次/只联合HBD2基因真核表达质粒pEGF-C1/HBD2 100μg/次/只,按间隔2周、共免疫3次的左后肢股四头肌肌肉接种方案免疫BALB/c小鼠,同时设立相应的对照。免疫后用双抗原夹心ELISA法测小鼠血清HBsAb水平。采用流式细胞仪检测脾脏CD4+T和CD8+T细胞百分率及CD4+/CD8+比值。应用CCK8试剂盒检测脾淋巴细胞在不同抗原刺激下的增殖反应。结果:pEGF-C1/HBD2质粒联合乙肝疫苗免疫小鼠抗体水平明显升高,CD4+T细胞百分率比其他免疫组都高,脾淋巴细胞体外增殖实验明显。与其他各免疫组相比,pEGF-C1空质粒联合乙肝疫苗免疫组抗体水平和淋巴细胞体外增殖情况及CD4+T细胞百分率都较低。结论:人β-防御素-2对乙肝疫苗在小鼠中的免疫功能有一定增强作用。Objective:To explore whether the beta-defensin-2 can strengthen the immune effectives of hepatitis B vaccine. Methods: Mice were immunized with both 2 μg hepatitis B vaccine and 100 μg pEGF-C1/HBD2 containing I-IBD2 eukaryotic expression plasmid by intramuscular injection in the left tibialis anterior muscle, and this kind of immunization was carried on 3 times by 2 weeks interval. The corresponding control group was set up at the same time. Serum antibody level in mice were measured by sandwich ELISA, and CD4 + and CD8 + T cell percentage of spleen and ratio of CD4 +/CD8 + were measured by flow cytometry, and the proliferation of spleen lymphocytes was tested by CCK8 assay under the stimulation from ConA or hepatitis B vaccine in vitro. Results:The lev- el of serum HBsAb from the hepatitis B vaccine combined with pEGF-C1/HBD2 immunized mice was higher than other groups. The proliferation in vitro and the CD4 + T cell percentage of spleen lymphocytes from the mice immunized with the plasmid pEGF-C1/HBD2 and hepatitis B vaccine were also higher than other groups. And all these figures from the group which was immunized with the empty plasmid pEGF-C1 plus hepatitis B vaccine were lower than others. Concluslon:Beta-defensin-2 may play a strengthen role of immunization of hepatitis B vaccine.
