摘  要:目的探讨丹酚酸B对动脉粥样硬化CD40-CD40配体信号通路的影响。方法 24只日本大耳白兔给予普通颗粒饲料喂饲1周后,随机分为三组,每组8只:正常对照组、高脂模型组、丹酚酸B组,后两组家兔分别通过高脂饮食制备动脉粥样硬化模型。于给药12周末处死各组实验兔,获取血浆和主动脉全长标本。比较血脂、主动脉病理形态学改变,免疫组织化学法测定斑块部位CD40L的表达,采用酶联免疫吸附法测定可溶性血管细胞粘附分子-1和可溶性细胞间粘附分子-1变化,Western杂交分析测定基质金属蛋白酶-9的蛋白表达。结果与高脂模型组比较,丹酚酸B组兔的主动脉CD40L的表达降低(P<0.01),血清可溶性血管细胞粘附分子-1、可溶性细胞间粘附分子-1及基质金属蛋白酶-9的蛋白表达明显降低(P<O.01),且兔主动脉内膜/中膜厚度比值明显降低。结论丹酚酸B减轻动脉粥样硬化病变的作用,与抑制CD40-CD40配体信号通路,降低粘附分子及基质金属蛋白酶的表达水平有关。【Objective】 Explore the effects of Salvianolic acid B On CD40-CD40L signal pathway in atherosclerosis model.【Methods】 24 healthy Japanese big-ear rabbits were randomLy divided into three groups with 8 rabbits in each group:Normal control group 、Model group 、Salvianolic acid B group.The rabbits in model group and Salvianolic acid B group have been used to prepare the atherosclerosis model induced by high-cholesterol diet.At the end of the experiment,blood samples of the rabbits were collected for biochemistry analysis and aorta was prepared for morphologic analysis.【Resluts】as compared with the model group,The level of CD40L、sVCAM-1、sICAM-1 and MMP-9 have also decreased in danshen group with statistical significance(P0.01),ratios of intima /media thickness in Salvianolic acid B group have been reduced significantly(P0.01).【Conclusion】Salvianolic acid B reduce the role of atherosclerotic lesions,and inhibit CD40-CD40 ligands signaling pathways,reduce adhesion molecules and matrix metalloproteinases expression level of relevant.
