疏肝健脾方对腹泻型肠易激综合征大鼠5-羟色胺相关的结肠黏膜上皮分泌功能的影响

摘　　要：目的:探讨5-羟色胺(5-HT)对腹泻型肠易激综合征(IBS-D)大鼠及疏肝健脾方干预后IBS-D大鼠结肠黏膜上皮分泌功能的影响。方法:将SD大鼠随机分成正常组、模型组、疏肝健脾组,模型组及疏肝健脾组采用乙酸灌肠加束缚应激进行造模,造模成功后,正常组和模型组予0.9%氯化钠溶液灌胃,疏肝健脾组予疏肝健脾方灌胃。采用恒温灌流装置体外测量跨上皮电流为短路电流。观察5-HT、去甲肾上腺素(NE)对各组大鼠结肠上皮短路电流的影响。结果:模型组大鼠基础电流较正常组及疏肝健脾组高,但是差异无统计学意义;5-HT引起的模型组大鼠结肠黏膜上皮产生的短路电流较正常组及疏肝健脾组大鼠降低(P<0.05);去甲肾上腺素引起的模型组大鼠结肠黏膜上皮产生的短路电流降低幅度较正常组及疏肝健脾组低。结论:5-HT引起的IBS-D大鼠结肠黏膜上皮的分泌活动较正常大鼠弱,疏肝健脾方通过改善IBS-D大鼠对5-HT的反应性达到治疗目的。Objective： To investigate the effect of 5-HT as well as Shu-Gan-Jian-Pi Prescription（SGJP） on the ion transportation of IBS-D colon mucosa.Methods： Male SD rats were randomLy divided into three groups： normal group,IBS-D group and SGJP group.IBS-D Rat model was induced by intracolonic instillation of acetic acid and restraint stress.The dosage for SGJP group rats was 3.4g/100g,the normal group and IBS-D group were given equivalent volume of 0.9% NaCl,all animals were intragastrically administered for 14 days after modeling,then the colon segments were collected for short circuit current test with bathing instrument in vitro.Observe the 5-HT and NE on the shirt circuit current change of each group.Results： The distal colon of IBS-D,normal and SGJP group showed no difference in basal curren t（BC）;5-HT induced short circuit current of IBS-D group significantly decrease compared to normal and SGJP group（P0.05）;NE induced short circuit current of IBS-D group significantly decrease compared to normal and SGJP group.Conclusion： Compared to the normal group,the ion absorption induced by 5-HT in IBS-D group significantly decreased,but this response recovered after treatment with SGJP.
