补肾健脾方对大鼠脾肾两虚型骨质疏松症的治疗作用

摘　　要：目的:深入探讨补肾健脾方对脾肾两虚型大鼠骨质疏松症的治疗作用,并为骨质疏松发生过程中脾肾相关性研究提供实验依据。方法:建立脾肾两虚型大鼠骨质疏松模型,并在此基础上,分别给予阳性对照药、补肾方药、补脾方药及补肾健脾方药进行治疗。给药3个月结束后,采用骨组织形态计量学方法检测反映骨量的指标骨小梁体积百分比(TBV%),反映骨吸收情况的指标骨小梁吸收表面百分比(TRS%)以及反映骨形成情况的指标骨小梁形成表面百分比(TFS%)、类骨质平均宽度(OSW)、骨小梁矿化率(MAR)和骨皮质矿化率(mAR)。结果:与假手术组相比,脾肾两虚模型组大鼠胫骨TBV%显著降低(P<0.01),TRS%、TFS%、OSW、MAR和mAR显著增高(P<0.01)。与脾肾两虚模型组相比,补肾组、健脾组、补肾健脾组、己烯雌酚组TBV%均显著增高(P<0.05,P<0.01),TRS%、TFS%、OSW、MAR和mAR均显著降低(P<0.01)。与健脾组相比,补肾组及补肾健脾组的TBV%明显增高(P<0.05,P<0.01),TRS%和TFS%明显降低(P<0.05,P<0.01);且补肾健脾组对上述指标的逆转较补肾组更为显著(P<0.05,P<0.01)。结论:健脾法、补肾法和补肾健脾法对脾肾两虚型大鼠骨质疏松症均具有治疗作用,其中补肾健脾法的效果最佳。提示在骨质疏松症的发生过程中,中医的脾肾两脏是密切相关的。Objective： Investigate deeply the therapeutic effect of recuperating kidney and invigorating spleen prescription on rats with spleen and kidney deficiency syndrome to provide experimental basis for the correlation study of spleen and kidney in the process of osteoporosis occurrence.Methods： Establish a rat osteoporosis model with spleen and kidney deficiency syndrome.On this basis,treat the model rats respectively with prescriptions of positive control,recuperating kidney,invigorating spleen,recuperating kidney and invigorating spleen.After three months administration,measure the indexes of TBV%,TRS%,TFS%,OSW,MAR and mAR,which can reflect the bone mass,bone formation and bone resorption.Results： Compared with the sham operation group,TBV% in spleen and kidney deficiency model group decreased significantly（P0.01）,TRS%,TFS%,OSW,MAR and mAR increased significantly（P0.01）.Compared with the model group,TBV% in all treatment groups increased obviously（P0.05,P0.01）,TRS%,TFS%,OSW,MAR and mAR decreased obviously（P0.01）.Compared with invigorating spleen group,TBV% increased and TRS%,TFS% decreased obviously in recuperating kidney group and recuperating kidney and invigorating spleen group（P0.05,P0.01）,and the later reversed these indicators more significant（P0.05,P0.01）.Conclusion： The methods of recuperating kidney,invigorating spleen,recuperating kidney and invigorating spleen have therapeutic effects on rats with spleen and kidney deficiency syndrome,and the methods of recuperating kidney and invigorating spleen work best.It points that in the process of osteoporosis,spleen and kidney of Chinese medicine are closely related.
