摘 要:目的:探讨黄芪多糖联合丹参酮阻断慢性心力衰竭(CHF)大鼠核因子-κB(NF-κB)激活巨噬细胞移动抑制因子(MIF)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)表达的效应。方法:应用大鼠左冠状动脉结扎术建立心肌梗死后心衰模型,第4周行超声多普勒心电图检查,将30只成模大鼠随机分6组:模型组、黄芪多糖组、丹参酮组、黄+丹组、他汀类药物组、假手术组,灌胃治疗6周后制备病理切片,SP法免疫组化染色,RT-PCR法检测心肌组织NF-κB mRNA、MIF mRNA表达,ELISA法检测血清MIF、TNF-α、IL-6含量。结果:模型组心肌细胞减少,成纤维细胞增殖,炎细胞浸润(P<0.05);模型组NF-κB、MIF阳性染色率及NF-κB mRNA、MIF mRNA表达均明显高于假手术组(P<0.05);模型组血清MIF、TNF-α、IL-6含量明显高于假手术组,治疗组能明显抑制血清各细胞因子表达,黄+丹组与黄芪多糖组及丹参酮组相比,具有显著性差异(P<0.05)。结论:黄芪多糖联合丹参酮对慢性心衰心肌各细胞因子的表达具有重要的抑制效应,阻断NF-κB信号通路可能对于慢性心衰心肌损伤具有潜在治疗价值。Objective: To investigate Astragalan combined with Tanshinone blocking nuclear factor-κB(NF-κB) activation on the expression of macrophage migration inhibitory factor(MIF),tumor necrosis factor-α(TNF-α),interleukin-6(IL-6) in rats with chronic heart failure.Methods: The heart failure model after myocardial infarction caused by left coronary artery ligation in rats was established.Echocardiography was used in the fourth week,and model rats were divided into 6 experimental groups: model group,Astragalan group,Tanshinone group,Astragalan combined with Tanshinone group,Atorvastatin group and sham group.Pathological section was made and SP immunohistochemical staining was used.RT-PCR was used to detect myocardial NF-κB mRNA,MIF mRNA expression,and ELISA was used to test serum MIF,TNF-α and IL-6 levels.Results: After 6 weeks,the left ventricular histopathology analysis showed a decrease of myocardial cells,an increase of fibroblasts proliferation and inflammatory cells infiltration in the model group(P0.05);RT-PCR assay suggested that the expression of NF-κB mRNA and MIFmRNA of the model group were significantly higher than the sham group(P0.05);ELISA method results suggested that the levels of serum MIF,TNF-α and IL-6 of the model group were significantly higher than the sham group,the treatment could significantly inhibit the serum cytokines.Compared with other experimental groups,compatibility group was significantly different(P0.05).Conclusion: Astragalan combined with Tanshinone has an important inhibitory effection on the expression of each cytokine in chronic heart failure.Blocking NF-κB signaling pathway for chronic heart failure may have potential therapeutic value.
