黄芪穴位注射对运动性疲劳大鼠下丘脑—垂体—肾上腺轴的影响

摘　　要：目的分析黄芪穴位注射对运动性疲劳大鼠HPA轴的影响。方法将32只SD雄性大鼠随机分为正常组、模型组、黄芪穴位注射组和生理盐水穴位注射组。黄芪穴位注射组予以足三里穴位注射黄芪注射液0.1 mL。生理盐水穴位注射组穴位注射0.1 mL生理盐水。每天游泳后称量体重。末次游泳结束后,穴位注射1 h后,水合氯醛麻醉,心脏取血后,取下丘脑,用生理盐水制备20%匀浆。检测下丘脑匀浆中促皮质激素释放激素(CRH)以及血清中促肾上腺皮质激素(ACTH)、皮质酮(CORT)的含量。结果模型组下丘脑CRH含量较正常组有降低趋势,血清CORT含量显著低于正常组;黄芪穴位注射组大鼠下丘脑CRH含量较模型组略有增高,血清ACTH、CORT含量均明显低于模型组。结论黄芪穴位注射可以减少疲劳大鼠的体重下降,可以促进大鼠下丘脑分泌CRH,抑制ACTH和CORT的分泌,减弱HPA轴功能亢进引起的疲劳反应。Objective To analyze the impacts on HPA axis in the rats with sport fatigue treated with acupoint injection of radix astragali.Methods 32 SD male rats were randomized into a normal group,a model group,a radix astragali injection group and a physiological saline injection group.In the radix astragali injection group,the radix astragali injection 0.1ml was applied to Zusanli（ST36）.In the physical saline injection group,the physiological saline injection 0.1ml was used.After swimming,the body weight was measured every day.At the end of the last swimming,1h after acupoint injection,under the anesthesia with chloral hydrate,after the heart collected,the hypothalamus was removed and mixed with 20% homogenate was prepared with physiological saline.The contents of corticotropin releasing hormone（CRH）in hypothalamus homogenate,and adrenocorticotropic hormone（ACTH）and corticosterone（CORT）in serum were detected.Results The content of hypothalamus CRH in the model group was reduced in tendency compared with that in the normal group,the content of serum CORT was lower significantly than that in the normal group.In the radix astragali injection group,the content of hypothalamus CRH in the rats were increased slightly as compared with that in the model group,the contents of serum ACTH and CORT were lower obviously than those in the model group.Conclusion Acupoint injection of radix astragali reduces the decrease of body weight,promotes the secretion of hypothalamus CRH,inhibits the secretion of ACTH and CORT and alleviates fatigue reactions induced by the hyperactivity of HPA axis in rats.
