摘 要:目的:制备苦豆子总碱缓释磁性颗粒,并研究其性能。方法:采用L9(34)正交试验确定苦豆子总碱缓释磁性颗粒最佳制备工艺。以pH6.8的磷酸盐缓冲溶液为释放介质,考察其体外释放性能,并通过振动样品磁强计测定其磁性能。结果:苦豆子总碱缓释磁性颗粒最佳制备工艺组成为A1B3C2D2,即苦豆子总碱质量为1.0g,壳聚糖浓度为2.5%,壳聚糖质量为3.0g,微晶纤维素质量为1.5 g,Fe3O4纳米粒质量为1.5 g。苦豆子总碱缓释磁性颗粒具有较好的缓释性能,其体外释药特征符合一级动力学方程,同时也具有明显的磁响应性。结论:苦豆子总碱缓释磁性颗粒处方合理、制备工艺可行,具有缓释特性和超顺磁性,适用于消化道靶向给药。OBJECrIVE To prepare Sophora alopecuroides L. total alkaloids sustained release magnetic granules (SATA SRMG), and study its characterization. METHODS The formulation of the SATA-SRMG was optimized by orthogonal design of L9 (34). Using phosphate buffer solution (pH 6. 8) as release solvent,the delivery property in vitro release was determinedby HPLC via rotating basket method. Also, the magnetic property was determined with vibrating sample magnetometry. RE SULTS The optimized preparation technology for this granules were belowed as follows: Sophora alopecuroides L. total alka loids 1.0 g, 2. 5% chitosan 3.0 g, microcrystalline cellulose 1.5 g, Fe3 O4 nanoparticle 1.5 g, i. e. A1 B3 C2 D2. The release mechanism of SATA-SRMG in vitro may be described through first-class kinetics affiliated sustained- release feature. In addi tion, hysteresis curve suggests that the SATA-SRMG possessed satisfactory time-independent magnetic responsive ability. CONCLUSION The preparation composition were rationable for the SATA-SRMG, and the preparation method of SATA SRMG is reasonable and practicable. It may be refer to target delivery in alimentary tract.
