不同时点应用甘松挥发油对大鼠心肌缺血再灌注损伤的影响

摘　　要：目的:观察不同时点应用甘松挥发油对大鼠心肌缺血再灌注损伤的影响。方法:将SD大鼠40只随机分成4组,每组10只:(1)假手术组;(2)心肌缺血再灌注(I/R)组;(3)甘松挥发油-1组;(4)甘松挥发油-2组。分别观察各组大鼠心肌梗死面积、血清CK-MB和cTnT水平,以及血清IL-6和TNF-α含量;并使用心电图记录再灌注期间室性早搏频数和室速持续时间。结果:甘松挥发油-1组和甘松挥发油-2组中心肌梗死面积、CK-MB、cTnT、IL-6、TNF-α均明显小于I/R模型(P均<0.05),其中甘松挥发油-1组中心肌梗死面积、CK-MB、cTnT、IL-6、TNF-α均明显小于甘松挥发油-2组(P<0.05)。结论:在大鼠心肌缺血再灌注前使用甘松挥发油进行干预能显著减轻心肌损伤,其机制可能与甘松挥发油具有钙通道阻滞剂作用、抑制炎症反应、延长心肌细胞有效不应期有关。OBJECTIVE To investigate the effects of the volatile oil of Nardostachys chinensis administered at different time points on myocardial ischemia-reperfusion injury in rats. METHODS Forty Sprague Dawley rats were randomly divided into sham group（n = 10）, ischemia/reperfusion group （n = 10） , the volatile oil of Nardostachys chinensis group-t （n = 10）; the vob atile oil of Nardostachys chinensis group-2（n = 10）. The infarct size , serum levels of interleukin-6 （IL-6） and tumor necrosis factor-α （TNF-α）,CK-MB,eTnT in each group were investigated; and the ventricular arrhythmias in each group were recor ded. RESULTS The serum levels of CK-MB, cTnT, IL-6, TNF-α and infarct size were significantly reduced in the volatile oil of Nardostachys chinensis group-I and the volatile oil of Nardostachys chinensis group-2 （P〈0. 05）, however the reduction was more significant in the volatile oil of Nardostachys chinensis group-1. CONCLUSION The volatile oil of Nardostachys chinensis administered before myocardial ischemia-reperfusion iniury in rats had protective effects on myocardial ischemia/reper fusion injury. The mechanism of action may possibly relate to the inhibi!ing early inflammatory reaction, blocking the L- type calcium channel , and the prolongation of the effective refractory period （ERP） of the heart muscle.
