摘 要:目的研究芪参复康胶囊对心理应激促发的高原低压低氧环境下大鼠肺损伤的干预效果。方法雄性SD大鼠随机分为低氧组,实验组(低氧+应激)和芪参复康胶囊干预组(低氧+应激+药物)。应激采用旁观电击心理应激模型。模拟高原低压低氧环境处置,把大鼠置于模拟海拔6000m低压舱内24h。用推挽灌流的方法收集海马细胞外液,用高效液相色谱-电化学测定海马细胞外液中的去甲肾上腺素(NE)的含量。通过测定大鼠肺湿/干比值、肺湿重/体重比值和与细支气管伴行小血管"袖套"面积/血管面积的比值来评定肺损伤程度。结果①实验组海马细胞外液中的NE含量、肺湿/干比值和血管"袖套"/血管比值均显著高于低氧组(t=2.453,2.514,2.813;P均<0.05);②干预组海马细胞外液中的NE含量、肺湿/干比值和血管"袖套"/血管比值均显著低于实验组(t=2.183,2.171,2.242;P均<0.05)。结论芪参复康胶囊能有效预防心理应激促发的低压低氧环境下的大鼠肺损伤。Objective To study the intervention effect of Qishenfukang capsules on lung injuryof rats under high altitude hypobaric hypoxia induced by psychological stress.Methods Male SD rats were randomly divided into three groups:Hypoxia group,experiment group(Psychological stress + hypoxia group),intervention group(Psychological stress + hypoxia+Qishenfukang capsules group).Psychological stress was induced the way of communication box model.The acute hypobaric hypoxia disposure was made by putting the rats into hypobaric chamber at a simulated altitude of 6000 meters for 24 hours.The extracellular fluid of hippocampus was collected by the push-pull perfusion method.The contents of norepinephrine(NE)in the extracellular fluid of hippocampus were determined by high performance liquid chromatography with electrochemical detection(HPLC-ECD).To assess lung injury,the wet weight/dry weight ratio of right lung,the wet lung weight /body weight ratio were determined.To further assess the lung injury,the perivascular cuff area of small vessels accompanied with bronchiole and the perivascular cuff area/ vessels area ratio were measured and calculated by using image analyzer.Results ①The contents of NE,wet weight/dry weight ratio of right lung and perivascular cuff area/ vessels area ratio in experiment group were significanly higher than in hypoxia group(t=2.453,2.514,2.813;P all0.05).②The contents of NE,wet weight/dry weight ratio of right lung and perivascular cuff area/ vessels area ratio in intervention group were significanly lower than in experiment group(t=2.183,2.171,2.242;P all0.05).Conclusion Qishenfukang capsules can utility prevent psychological stress promoting lung injury induced by simulated acute high altitude.
