摘  要:目的:用分子标记技术对中药溪黄草的基原植物分类鉴定。方法:用离心柱法提取5种溪黄草基原植物基因组DNA,用85个10 bp的寡核苷酸随机引物进行PCR(聚合酶链式反应),将扩增产物进行琼脂糖凝胶电泳,分析电泳图谱。结果:从85条随机引物中筛选出了12条多态性较好的引物,其中7个引物的扩增图谱表现较好的多态性,且重复性较好。结论:RAPD技术可以成功鉴定溪黄草药材基原。Objective:To identify the different plant resources of Herba Rabdosiae Serrae by using Random Amplified Polymorphic DNA ( RAPD ) Analysis. Methods : The Mini Spin Columns were used to extract the genomic DNA from five different plants of Herba Rabdosiae Serrae. With the DNA extracted from these plants as template ,the 85 oligo nucleic acids (10 bp)as random primers ,the PoX- ymer Chain Reaction(PCR) was done and the results were analysed by electro-pharoses. Results: 12 primers were selected with poly- morphism and 7 of them showed good polymorphism in RAPD map. Conclusion: RAPD method can be used to identify the plant re- sources of Herba Rabdosiae Serrae.
