摘 要:目的:观察正常、超重和肥胖3组人群皮下和大网膜脂肪组织中维生素D受体(vitamin D receptor,VDR)mRNA及其蛋白表达水平的差异。方法:用半定量RT-PCR方法检测20例正常组、35例超重组、27例肥胖组人群的腹部皮下和大网膜脂肪组织VDR基因的mRNA表达水平;用Western blot方法检测VDR的蛋白表达水平。结果:在皮下脂肪组织中,肥胖组VDRmRNA和蛋白表达水平明显低于正常组和超重组(P<0.05);在大网膜脂肪组织中,超重组VDR mRNA及其蛋白表达增高(P<0.05),肥胖组VDR mRNA及其蛋白表达与超重组相比降低(P<0.05)。结论:肥胖患者脂肪组织中VDR表达水平降低,可能是临床上补充维生素D干预肥胖达不到预期效果的机制之一。Objective:To investigate the mRNA and protein expression differences of vitamin D receptor (VDR) in subcutaneous and omental adipose tissue of people with different BMI. Methods:Twenty people of normal BMI, 35 people of overweight and 27 people of obesity were selected. The mRNA transcription levels of VDR were assayed by RT-PCR, and the VDR protein expression was measured by Western blot. Results: ① In subcutaneous obesity tissue, mRNA transcription and protein expression of VDR was lower in obesity group than in the normal or overweight group (P 〈 0.05) ; ② In omental adipose tissue,mRNA transcription and protein expression of VDR was highest in overweight group among 3 groups (P 〈 0.05). Compared with the overweight group, mRNA transcription and protein expression of VDR was lower in obesity group(P 〈 0.05). Conclusion:The VDR expression decreased in the obesity group, which may be one of the mechanisms of the poor clinical outcome of vitamin D intervention therapy.
