摘 要:目的比较研究两种重要的丹酚酸化合物(丹酚酸A和丹酚酸B)的体外抗氧化活性。方法利用DPPH自由基清除、超氧阴离子清除、抗脂质过氧化、还原能力以及抑制脱氧核糖降解等体外实验,综合评价抗氧化能力。结果丹酚酸B(SAB)的电子传递和超氧阴离子清除能力强于丹酚酸A(SAA);SAA在DPPH自由基清除、抗脂质过氧化以及抑制脱氧核糖降解方面的活性均强于SAB,特别对于羟自由基定位介导的脱氧核糖降解具有显著的抑制作用,提示其为较强的金属离子络合剂。结论 SAA是一种极具开发价值的天然抗氧剂候选化合物,可能通过自由基清除、抗脂质过氧化、抑制脱氧核糖降解以及金属离子螯合发挥抗氧化作用。Objective To compare in vitro antioxidant activities of two important natural salvianolie acids, salvianolie acid A (SAA) and B ( SAB ). Methods Various in vitro measurements were applied, including radical scavenging rate of 1,1 - Diphenyl - 2 - pierylhydrazyl (DPPH) and O2^ - , Fe^3 + reducing power, inhibitory effects on liposome peroxidation and deoxyribose degra- dation. Results SAB was more effective than SAA in superoxide anion scavenging and electron donating activity. However, SAA displayed more potent antioxidant power in DPPH scavenging, and inhibition of liposome peroxidation and deoxyribose degradation. Moreover,relatively higher inhibitory effect of SAA on deoxyribose degradation in the site- specific model indicated that SAA was a potent chelating agent. Conclusion SAA is a naturally oecuring antioxidant with the great potential to be developed. Its antioxidative mechanism may be involved in free radical scavenging activity, chelating with metal ion, and inhibitory effects on liposome peroxidation and deoxyribose degradation.
