摘 要:目的从逍遥散复方提取分离得到的不同极性化学部位中,筛选出抗抑郁活性部位。方法①分别采用D101大孔吸附树脂柱分离和溶剂萃取法分离,通过HPLC图谱评价两种分离方法的分离效果。②以小鼠行为绝望模型评价逍遥散各个不同极性部位的抗抑郁作用。结果柱层析法得到的各部位HPLC图谱交叉较少,溶剂萃取法分离得到的各部位HPLC图谱交叉较多;有两个部位可缩短绝望模型中小鼠强迫游泳的不动时间(P<0.05),其中以最小极性部位最为显著(P<0.01)。结论在两种分离方法中,柱分离效果较理想,逍遥散抗抑郁作用的活性部位为极性较小的部位,为逍遥散抗抑郁药效物质基础研究奠定了基础。Objective To obtain different polar fractions of Xiaoyaosan and screen the antidepressant fractions. Methods Column chromatography and solvent extraction were used to separate Xiaoyaosan into different polar fractions. HPLC was used to evaluate the effect of two methods. Behavioral despair model of mice was used to evaluate antidepressant activities of the different polar fractions which were obtained by the preferable separation method. Results Different polar fractions obtained by column chromatography overlaped each other in HPLC fingerprint were less than solvent extraction. Two fractions were able to elicit time - dependent reduction in immobility duration in the forced swimming test in mice (P 〈 0. 05). Among these test samples, the lowest polar fraction from Xiaoyaosan was more effective ( P 〈 0. 01 ). Conclusion The separation effect of column chromatography is better than solvent extration. The antidepressant active fractions of Xiaoyaosan are low - polar. It laid the foundation for the study of antidepressant material basis of Xiaoyaosan.
