摘  要:目的探讨罗布麻叶总黄酮提取物的抗抑郁活性及其作用机制。方法采用小鼠悬尾实验及皮质酮损伤的PC细胞实验考察罗布麻叶总黄酮的抗抑郁活性及作用机制。结果给小鼠罗布麻叶总黄酮提取物25,50,100mg/kg后,小鼠悬尾累计不动时间显著缩短。罗布麻叶总黄酮提取物25,50,100μg/ml与皮质酮0.01mmol·L-1共孵PC12细胞48 h,可防止皮质酮所致的PC12神经细胞损伤,降低胞内Ca2+超载。结论罗布麻叶总黄酮提取物对小鼠悬尾模型有抗抑郁作用,其机理可能与神经细胞的保护作用及降低胞内Ca2+超载有关。Objective To explore the antidepressant effect and mechanism of flavonoids extracted from Apocymum venetum leaves. Methods The classical animal models of depression including the tail suspension of the mouse was applied to evaluate the anti- depressant effect of AV - extract, and neuroprotective effect of AV - extract was studied in corticosterone - induced neurotoxicity. Results The results showed that administration of AVextract(25,50 and 100 μg/ml, i. p. ) reduced the duration time of immobil- ity significantly. After incubation with the PC12 cells in the presence of corticosterone (0.01 mmol . L^-1 ) for 48 h, AV - extract decreased Ca^2+ concentration and protected PC12 cells from the injury by corticosterone. Conclusion The results suggested that AV - extract showed antidepressant - like effect in TST model and the possible mechanism may be related to the neuroprotection achieved by decreasing Ca^2+ concentration to promote cell proliferation
