肺岩宁方对肺癌干细胞Wnt信号通路的影响

摘　　要：目的:分离、鉴定肺癌干细胞,并探讨肺岩宁方对肺癌干细胞Wnt信号通路的干预作用。方法:分离、鉴定肺癌干细胞;将分离出的SP+肺癌干细胞分为对照组(空白血清+生理盐水)、单纯化疗(空白血清+DDP组)、肺岩宁方组(肺岩宁方含药血清+生理盐水)、综合治疗组(肺岩宁方含药血清+DDP),予体外药物干预后检测β-catenin蛋白表达及SP+肺癌干细胞凋亡情况。结果:分离的SP+、CD24+IGF-1R+、CD133+细胞具有肺癌干细胞特性,分离的β-catenin蛋白完整;综合治疗组诱导SP+干细胞凋亡优于其他组,各组凋亡率分别为综合治疗组79.2%、肺岩宁方组21.3%、DDP组33.7%、对照组0.3%。结论:本课题组已掌握分离、鉴定肺癌干细胞的技术,中药肺岩宁方联合化疗具有一定的诱导肺癌干细胞凋亡的作用。Objective： To separate and identify the lung cancer stem cells and explore the intervention of ＂ Feiyanning Decoction＂ on Writ signal pathway of lung caner stem cell. Methods： The lung cancer stem cells were separated and identified. The SP ＋ lung cancer stem cells were divided into control group （ treated with serum and saline）, chemotherapy group （ treated with serum and DDP）, ＂ Feiyanning Decoction＂ group （ treated with serum containing ＂ Feiyanning Decoction＂ and saline ）, combined therapy group （treated with serum containing ＂Feiyanning Decoction＂ and DDP）. The expression of β-catenin and the apoptosis of SP＋ lung cancer stem ceils were detected. Results： The separated SP＋ , CD24 ＋ IGF-1R＋ and CD133 ＋ lung cancer cells had the characteristics of lung cancer stem cells, and the β-catenin protein was integrity. The apoptosis of SP ＋ lung cancer ceils in the combined therapy group was better than that in the other groups, and the apoptosis rates of combined therapy group, ＂Feiyanning Decoction＂ group, chemotherapy group and control group were 79.2%, 21.3%, 33.7% and 0.3% respectively. Conclusion ： The technology of isolating and identifying the lung cancer stem ceils has been obtained, and ＂Feiyanning Decoction＂ combined with chemotherapy can induce the apoptosis of lung cancer stem cells.
