冠心舒通胶囊对慢性心力衰竭大鼠心肌组织MMP-9的影响

摘　　要：目的:观察冠心舒通胶囊对慢性心力衰竭大鼠心肌组织基质金属蛋白酶(MMP-9)的影响。方法:采用冠状动脉结扎法结合力竭式游泳、减食等方法复制大鼠慢性心力衰竭模型。将大鼠分为模型组、假手术组、西药组、益气活血组、冠心舒通组;采用SABC免疫组化法检测各组大鼠心肌组织MMP-9的变化情况。结果:与模型组比较,假手术组、西药组、益气活血组、冠心舒通组大鼠心肌组织MMP-9 mRNA、蛋白表达均明显降低(P<0.05);与假手术组比较,西药组、益气活血组、冠心舒通组大鼠心肌组织MMP-9mRNA、蛋白表达均明显升高(P<0.05);且3个治疗组之间无显著差异(P>0.05)。结论:冠心舒通胶囊可通过抑制心肌组织MMP-9表达以治疗慢性心力衰竭。Objective： To observe the effects of ＂Guanxin Shutong Capsule＂ on matrix metallopeptidase-9 （MMP-9） in the rats with chronic heart failure（CHF）. Methods： Chronic heart failure was duplicated by ligating the coronary artery, physical exhaustion and diet reduction. The rats were divided into five groups： model group, sham group, control group, positive group and treatment group, and treated with corresponding medicines. SABC immunohistochemical method was adopted to detect the expression of cardiac MMP-9 in all groups. Results： Compared with the model group, the MMP-9 expressions were decreased obviously in the other four groups（ P 〈 0.05 ） ; compared with the sham group, the MMP-9 expressions were elevated obviously in the other four groups（P 〈0.05 ） ; there was no difference in the MMP-9 expression between the control group, positive group and treatment group（P 〉0.05 ）. Conclusion： ＂Guanxin Shutong Capsule＂ is effective for chronic heart failure by reducing cardiac MMP-9 expression.
