摘 要:目的:探讨祛风止动方对抽动障碍(TD)大鼠纹状体神经递质基因表达的影响。方法:按照SPSS程序将SD大鼠随机分为正常组、模型组、中药大剂量组、中药常规剂量组及西药对照组,采用阿扑吗啡(APO)腹腔注射给药建立TD大鼠模型。中药组灌胃祛风止动方、西药组灌胃氟哌啶醇,正常组、模型组均予同等剂量生理盐水。药物干预4周后,断头处死实验大鼠,解剖脑组织并分离纹状体,采用RT-PCR检测各组大鼠纹状体中多巴胺受体D1/D2(DRD1/DRD2)mRNA表达水平。结果:腹腔注射APO后,经刻板行为验证显示模型大鼠能再现TD的特征性行为变化,且3周后模型组大鼠纹状体DRD2 mRNA明显上调、DRD1 mRNA明显下调,与正常组比较差异均有显著意义(P<0.05);经4周药物干预,西药组、中药大剂量组可显著上调DRD1 mRNA表达,明显优于中药常规剂量组(P<0.05);中药大剂量组可显著下调DRD2mRNA表达,明显优于西药组与中药常规剂量组(P<0.05)。结论:祛风止动方可调节纹状体的DRD1/DRD2 mRNA表达水平。Objective To explore the effect of "Qufeng Zhidong Recipe" (QZR) on neurotransmitter gene expressions in corpus striatum of rats with tic disorder. Methods: The SD rats were randomly divided into normal group, model group, high dose Chinese medicine group, regular dose Chinese medicine group and western medicine control group by SPSS software, and the tic disorder ( TD ) model was established by intraperitoneal injection of apomorphine ( APO ). The Chinese medicine groups were treated with QZR, the western medicine control group was treated with haloperidol, and the normal and model groups were treated with normal saline. After four weeks of intervention, all the rats were sacrificed, and the corpus striatum was separated from brain tissue. The expressions of dopamine receptor D1/D2 (DRD1/DRD2) mRNA in corpus striatum were detected by Real-time PCR. Results: After intraperitoneal injection of APO, the model could reflect the behavior characteristic of TD through stereotyped behavior validation, and three weeks later, the expression of DRD2 mRNA in corpus striatum was higher in model group than that in nm'mal group ( P 〈 0.05 ), while the expression of DRDI mRNA was lower in model group than that in the normal group ( P 〈 0.05 ). After four weeks of drug intervention, the expression of DRD1 mRNA was obviously increased in western medicine control group and high dose Chinese medicine group compared with regular dose Chinese medicine group ( P 〈 0. 05 ) ; the expression of DRD2 mRNA was significantly decreased in high dose Chinese medicine group compared with western medicine control group and regular dose Chinese medicine group(P 〈 0.05 ). Conclusion: QZR can adjust the expressions of DRDI/DRD2 mRNA in corpus striatum of rats with tic disorder.
