摘　　要：目的研究饮食诱导肥胖大鼠G蛋白耦联（门控）内向整流钾离子通道（GIRK4）基因在脂肪组织中的表达，探讨其与肥胖的相关性。方法30只Sprague—Dawley大鼠随机分为两组：对照组（n=10），予以普通饲料饲喂；肥胖组（n=20），予以高脂肪、高热量饮食饲喂。8周后处死大鼠，提取两组的白色脂肪组织和棕色脂肪组织，应用Western印迹技术检测GIRK4蛋白表达。并测定两组大鼠血糖、甘油三酯、血清总胆固醇水平。结果（1）共15只大鼠人组肥胖组（体重大于对照组大鼠平均体重20％）。（2）肥胖组血清总胆固醇及甘油三酯水平高于对照组（P〈0．05），但血糖在两组问差异没有统计学意义（P〉0．05）。（3）GIRK4在两组大鼠白色脂肪及棕色脂肪组织中均有表达，肥胖组的表达水平低于对照组（P〈0．05）。结论GIRK4可能肥胖的发生、发展中发挥一定作用。Objective To investigate the expression of G protein-coupled inward rectifier K＋ channels 4 （ GIRK4 ） gene in white adipose tissue and brown adipose tissue of diet-induced obese rats, and its relationship with obesity. Methods 30 Sprague-Dawley （SD） rats were randomly divided into two groups ：control group（ n = 10） ,which were fed with normal diet, and obese group （n = 20）, which were fed with high-fat,high-sugar diet. After 8 weeks all the rats were killed, and the white adipose tissue and brown adipose tissue were separated. The expression of GIRK4 were measure by Western-blotting, and the blood glucose,total cholesterol and triglyceride were detected. Results （ 1 ） 15 rats were enrolled in the obese group,whose weight were 20% higher than average weight of control group. （2） The levels of serum total cholesterol and triglyceride in the obese group were higher than control group （ P 〈 0.05 ） , while the level of blood glucose in two groups had no significant difference （ P 〉 0.05 ）. （ 3 ） GIRK4 were expressed in both white adipose tissue and brown adipose tissue of the two groups, and the level of GIRK4 in obese group is significantly lower than that of control group （ P 〈 0.05 ）. Conclusion GIRK4 may paly a role in the occurrence and development of obesity.
