摘　　要：为探索利用转基因植物生产鸡马立克氏病疫苗的新途径，将马立克氏病病毒（Marek’sdiseasevirus，MDV）B抗原基因的两个片段（I3和K3）整合连接到相应的双元载体上，构建成带有NptⅡ和Gus基因的MDVB抗原基因的植物表达载体pArt69I3K3。利用农杆菌LBA4404介导，将构建的表达载体导入烟草，获得了经PCR扩增和Southern杂交证实的转基因植株。免疫斑点杂交显示，转MDVB抗原基因的烟草植株叶片蛋白质提取物能与MDV单克隆抗体BA4（MDVB抗原Ⅰ型单克隆抗体）、BD8（MDV抗原群共同性决定簇单克隆抗体）和鸡MDV抗血清产生阳性免疫反应，表明MDVB抗原基因在转基因烟草植株中得到初步表达。A plant expression vector pArt69I3K3 carrying chicken Marek's disease virus (MDV) B antigen gene was constructed by linking the gene to a constitutive promoter (CaM V 35S) and terminator (Ocs, 3'). Tobacco plants were transformed by using Agrobacterium tumefaciens strain LBA 4404 harboring pArt69I3K3. Marker genes NPT Ⅱ and GUS were expressed in the putative transformants. PCR and Southern blot analyses of the transformants showed that MDV B antigen gene was integrated into the genome of tobacco. Protein dot blot by using BA4 and BD8 monoclonal antibody of MDV B antigen indicated that MDV B antigen gene expressed in transgenic tobacco plants. This result suggests that there was no inherent limitation for expressing MDV B antigen gene in plants.
