摘 要:用花粉管通道法将新疆大赖草DNA导入普通春小麦花培品系761(主穗长9.5cm),在导入后代D3代中筛选出一个大穗、晚熟变异株,经常规选有获得大穗(平均主穗长15.2cm,50个系)、多粒的转化后代。以地高辛标记的4个大麦高度重复序列克隆为探针,对供体、受体和转化后代进行Southern杂交分析。结果表明转化后代中出现了受体没有而供体具有的特异带,证明新疆大赖革DNA片段已整合到受体基因组中。因此认为外源DNA直接导入技术是克服小麦与其近缘野生种间杂交不亲和性的一条可行途径。Lcmpusmcemosus DNAs were introduced into common spring wheat Pollen line 761 through Pollen-tube pathway, A large spike and late mature variant was selected in D, generation, and from this variant progeny with large-spike and more grains were obtained. By using four repetitive DNA sequences (pHv7161, pHv7179, pHv7191 and pHv7293 ) cloned from the barley (Hordeum vulgare ) as probes, the results of Southern hybridization showed that there existed some specific fragments from donor plants in the progeny of transformed plants, indicating Lcmpus racemosus DNA fragments were transfered into acceptor plants pollen line 761. It is thought DNA transfer tCchnology through Pollen-tube pathway is efficient in overcoming the cross-incompatibility between common wheat and its wide relatives.
