摘  要:以太湖鹅和罗曼鹅为研究素材,应用实时荧光定量PCR技术和ELISA技术,对MSTN基因在2个鹅种中的表达差异及其与骨骼肌生长的相关性进行比较研究。结果发现,2个鹅品种下丘脑、垂体、肝脏中MSTN基因基本都不表达,只在胸肌、腿肌中表达。罗曼鹅腿肌重显著高于太湖鹅,太湖鹅腿肌MSTN基因表达量却极显著高于罗曼鹅;腿肌率和胸肌MSTN蛋白表达量呈显著正相关,腹脂率和胸肌MSTN基因表达量呈显著正相关;表明MSTN基因发挥着抑制骨骼肌生长的作用,同时可以促进脂肪的积累。在2个鹅种中,公鹅腿肌MSTN蛋白表达量高于母鹅,表明70日龄时的仔鹅腿肌生长母鹅快于公鹅;腿肌MSTN基因表达量显著高于胸肌,提示70日龄时的仔鹅胸肌生长速度远远大于腿肌。The expressions of MSTN gene in the breast muscle and leg muscle of Taihu goose and Roman goose were detected by RT-PCR and ELISA technology. It was the first study of MSTN gene's effect on the skeletal muscle growth in these two goose breeds. The result showed that no expression of MSTN gene was detected in hypothalamus ,hypophysis and liver of Taihu goose and Roman goose. MSTN gene expressed in both chest and leg muscle of the two breeds. Roman goose had significant higher leg muscle weight than Taihu goose, but Taihu goose had significantly higher MSTN gene expression in leg muscle than Roman goose. Leg muscle rate and expression of MSTN gene in chest muscle were significantly positive correlated ; abdominal fat percentage was significant positively correlated with expression of MSTN gene in chest muscle. Results showed that goose MSTN gene had the anti-growth effect in skeletal muscle, and could promote fat storage. At 70 clays of age, MSTN protein expression level was significant higher in male goose,this indicated female goose grew faster; higher protein expression level of leg muscle indicated chest muscle grew faster than leg muscle.
