摘  要:目的研究出生前后双酚A(BPA)暴露对子鼠雄性生殖细胞凋亡的影响,探讨BPA对雄性生殖系统的影响及可能机制。方法受孕0天(PGD0)的ICR孕鼠随机分为空白对照组、溶剂对照组、10 nmol/L BPA组、100 nmol/L BPA组、1000 nmol/L BPA组;孕鼠饮水染毒,出生后6周(PND42)分别处死雄性仔鼠,取睾丸称湿重,计算睾丸脏器系数;取睾丸组织,电镜观察超微结构的改变,Hoechst染色观察细胞凋亡,免疫组化检测Caspase-3的表达。结果与空白组和溶剂对照组比较,BPA暴露组睾丸脏器系数降低;电镜显示,高、中剂量组的间质细胞、精原细胞、支持细胞线粒体呈空泡样变化,粗面内质网呈不同程度的扩张。Hoechst染色显示中、高剂量组较对照组和低剂量组细胞凋亡明显。免疫组化结果显示高、中剂量组Caspase-3表达明显,多分布在精原细胞和间质细胞中,且间质细胞强阳性。结论BPA可以通过诱导生精细胞和间质细胞的凋亡影响睾丸组织学结构。Objective To investigate the effects ofbisphenol A (BPA) on spermatogenic cell apoptosis of offspring mice prior and postnatal to birth, and explore the possible molecule mechanism of BPA treatment. Methods ICR pregnant mice (0 day) were randomly divided into control group, solvent group, 10 nmol/L, 100 nmol/L, and 1000 nmol/L BPA groups, respectively. Pregnant mice of BPA groups were feed with BPA treated water. Testicular tissues of male offspring mice were taken out for evaluating testicular index at the six weeks atter the birth. Ultrastructure of testicular tissue was observed under electron microscope, cell apoptosis was measured by Hoechst dyeing, and caspase-3 expression was detected by immunohistochemical method. Results Compared with control and solvent groups,testicular index decreased in all BPA groups. Under electron microscopy, mitochondria vacuole and rough endoplasmic reticulum expansion appeared in leydig's cells, spermatagonial cells and support cells of 100 nmol/L and 1000 nmol/L BPA groups. Hoechst dyeing showed that cell apoptosis was more significant in 100 nmol/L and 1000 nmol/L BPA groups than that in control and 10 nmol/L BPA group. Immunohistochemical results showed the level of caspase-3 was more high in 100 nmol/L and 1000 nmol/L BPA groups, which mainly appeared in spermatagonial cells, leydig's cells and the leydig's cells. Conclusions Bisphenol A affect histological structure of the testicles through inducing apoptosis of sperm cells and stromal cell.
