摘  要:目的研究大蒜油(garlic oil,GO)对正己烷(n-hexane)染毒大鼠CYP2E1、CYP2B1和ADH的影响及其机制。方法雄性Wistar大鼠50只,随机分为正常对照组、大蒜油对照组、正己烷模型组、正己烷+大蒜油低、高剂量组(n=10);模型组、大蒜油低、高剂量组大鼠分别给予2000 mg/kg·bw正己烷灌胃,大蒜油低、高剂量组大鼠于正己烷灌胃前1 h分别给予40和80 mg/kg·bw大蒜油灌胃,大蒜油对照组给予80 mg/kg·bw大蒜油灌胃,每周6次,持续10周。每2周测步态评分,监测大鼠周围神经损伤情况。10周末,检测肝组织中CYP2E1、CYP2B1和乙醇脱氢酶(ADH)表达及活性,以及血清中2,5-己二酮(2,5-HD)含量变化。结果与模型组相比,大蒜油低高剂量组步态评分分别降低了38.2%和41.2%(P<0.05),血清2,5-HD含量分别降低47.7%和78.7%(P<0.01);肝脏CYP2E1含量及活性分别降低了32.9%、39.1%和27.4%、44.5%(P<0.01);肝脏CYP2B1含量及活性高剂量组升高了57.2%和52.6%(P<0.01);肝脏ADH含量变化不明显,但活性显著降低了25.3%和86.0%(P<0.01)。结论大蒜油能保护正己烷所致的大鼠周围神经损伤,机制可能与肝脏CYP2E1、CYP2B1及ADH表达或活性改变,正己烷转化为2,5-HD减少有关。Objective To study the protective effect of garlic oil(GO) on the peripheral nerve injuries induced by n-hexane. Methods Fifty male Wistar rats were randomly divided into five groups (n = 10) : the control group, the GO control group, the n-hexane group, the low dose GO and the high dose GO groups. The rats in the low and high doses of GO groups were pretreated with GO (40 and 80 mg/kg.bw) before exposed to n-hexane (ig, 2000 mg/kg.bw) , while the animals of the n-hexane group received equal volume of saline and then the same dose of n-hexane. The GO control group rats were treated with GO (80 mg/kg, bw). The gait scores were determined every two weeks. The rats were sacrificed at the end of 10 weeks. The activities and protein levels of hepatic CYP2E1, CYP2B1, ADH and 2, 5-HD in serum were examined. Results Compared with the n-hexane group, the gait scores in low and high doses of GO groups increased significantly ( P 〈 0. 05 ) ; the contents of serum 2,5-HD of the low and high doses of GO groups decreased by 47.7% and 78.7% respectively; the protein levels of hepatic CYP2E1 decreased 32.9% , 27.4% ,and activities decreased 39.1% ,44.5% , respectively (P 〈 0. 01 ) ; While the protein levels of hepatic CYP2B1 increased 3.3% , 57.2% ,and activities increased 13.1% , 52.6% , respectively (P 〈 0.05, P 〈0. 01, P 〈0. 05, P 〈0. 01 ) , the protein levels of ADH were not changed, but the activities were decreased 25.3% and 86. 0% (P 〈 0. 01 ). Conclusion The administration of GO has dramatically neuroprotective effects against the development of n-hexane neuropathy, which may be related to the changes of CYP2E1, CYP2B1 and ADH.
