摘 要:目的建立浙贝乙素胆酸盐中浙贝乙素和胆酸的含量测定方法。方法以甲醇-乙酸铵(0.1 mol.L-1)-三乙胺-冰乙酸(78∶22∶0.2∶0.03)为流动相,C18柱(250 mm×4.6 mm,5μm)分离,流速为1 mL.min-1;蒸发光散射检测器检测(漂移管温度72.1℃,氮气流速2 mL.min-1)。结果浙贝乙素胆酸盐中浙贝乙素和胆酸分别在50.4~151.1μg.mL-1(r=0.999 6,n=5)、50.5~151.4μg.mL-1(r=0.999 9,n=5)内峰面积与浓度呈良好线性关系;平均回收率分别为99.17%(RSD=0.62%)和99.37%(RSD=0.63%)。结论该方法测定浙贝乙素胆酸盐中的浙贝乙素和胆酸含量简便,可靠,重复性好。Objective To establish an HPLC method for determination of verticinone and cholic acid in verticinone- cholic acid salt. Methods The separation was performed on a C18column (250 mm×4.6 mm,5 μm). The mobile phase was methanol : ammonium acetate (0.1 mol· L-l) : triethylamine : ethanoic acid (78: 22 : 0.2 : 0.03). The flow rate was 1 mL·min-. The gas flow rate of ELSD was 2 mL · min-1 and the temperature of drift tube was 72.1℃. Results The linear ranges of vertieinone and cholic acid were 50.4-151.1μg · mL-l( r = 0. 999 6, n = 5 ) and 50.5-151.4 μg · mL-l( r = 0. 999 9,n = 5) respectively. The average recoveries of the two components were 99.37% ( RSD = 0.63% ,n = 9) and 99.17% ( RSD = 0.62%, n = 9 ) respectively. Conclusion The method is simple and reliable for the quality control of verticinone and cholieacid salt.
