摘  要:从湖北省武汉市某鸭场送检的死亡鸭肝脾中分离到1株病毒,结合流行病学调查、剖检变化等初步诊断为鸭瘟病毒感染。根据在GenBank上已发表的鸭瘟病毒TK基因序列AY963569设计引物,提取病毒核酸进行PCR扩增,得到预期大小为1 077bp的目的片段,经序列分析比较,与发表的鸭瘟病毒TK基因序列同源性达到99.16%。动物回归试验中感染雏鸭出现鸭瘟的典型临床症状,在透射电镜下观察到胸腺细胞中有与鸭瘟病毒颗粒的形态相符的病毒颗粒,上述结果表明分离的病毒为鸭瘟病毒。In this research,one virus strain Was isolated from the liver and spleen of dead duck lings in a farm in Wuhan, Hubei Province. Combined with epidemiological investigations and pathological of the duck lings, it is suspected that the reason for the death of the duck was duck plague virus infection. According to the genome of duck plague virus TK gene highly conserved sequence which was published in GenBank, two pairs of primers were designed. By PCR amplification of viral nucleic acid extraction, we achieved the expected size of 1 077 target fragment. Sequence analysis and comparison demonstrated that the homology is 99.16% compared to published duck plague virus TK gene sequence. Infected ducklings appeared the typical clinical symptoms of duck plague disease. Viral particles consisted with duck plague virus particles were observed in the thymus cells under transmission electron microscope. The date showed that this inspection cases is duck plague virus infection.
