摘  要:目的检测维生素K2对肝癌细胞增殖及肝癌衍生生长因子表达的作用。方法采用3种肝癌源性细胞株(HepG2、HUH-7和SK.Hep-1)进行细胞增殖实验。噻唑蓝(MTF)比色法测定不同浓度的维生素(2(0、10、30、100ixmol/L)干预后肝癌细胞数量。以半定量逆转录一聚合酶链反应(RT—PCR)检测0、100μmol/L维生素1(2干预后3种肝癌源性细胞株(HepG2、HuH.7和SK—Hep-1)中的HDGF的mRNA表达。结果肝癌细胞株HepG2、HUH-7、SK—Hep-1细胞在10μmol/L浓度维生素K2组存活率分别为(88.3±4.8)%、(94.2±4.3)%、(82.2±6.6)%;30μmol/L浓度维生素I(2组存活率为(75.4±4.3)%、(63.7±3.5)%、(58.1±7.5)%;100μmol/L浓度维生素I(2组存活率为(53.0±4.0)%、(21.2±3.6)%、(25.3±6.3)%。维生素K2呈剂量依赖性抑制3个肝癌细胞株的生长。同时,维生素K2显著性抑制3种细胞株的HDGFmRNA的表达。结论维生素K2抑制肝癌细胞增殖,并抑制肝癌细胞内HDGF基因表达。Objective To investigated the inhibitory effects of Vitamin K2 on the proliferation of human hepatocellular carcinoma ceil ( HepG2, HUH-'/ and SK-Hep-1 ) and the expression of hepatoma-de- rived growth factor (HDGF). Methods The inhibitory effect of Vitamin K2 on the proliferation of HepG2, HUH-7 and SK-Hep-1 was measured by methyl thiazol tetrazolium (MTr) assay. Semi-quantitative reverse- transcription-polymerase chain reaction (RT-PCR) was used to measure the mRNA levels of HDGF in hu- man hepatoeellular carcinoma cell ( HepG2, HUH-7 and SK-Hep-1 ) after 96 hours exposure to 0, 100 txmo]/L of Vitamin K2. Results Human hepatocellular carcinoma cell number ( HepG2, HUH-7, and SK- Hep-1 ) survival in 10μmol/L Vitamin K2 was ( 88.3 ± 4.8 ) % , (94.2 ± 4.3 ) % , ( 82.2 ± 6.6 ) % ; in 30μmol/L Vitamin K2 was (75.4 ± 4. 3 ) %, (63.7 ± 3.5 ) %, ( 58. 1 ± 7.5 ) % ; in 100μmol/L Vita- min K2 was (53.0 ±4. 0)%, (21.2 ±3.6)%, (25.3 ±6. 3)%. Vitamin K2 supresses the proliferation of human hepatoce|lular carcinoma cell in dose-dependent manner. By RT-PCR, Significant suppression of the HDGF mRNA expression in Human Hepatocellular Carcinoma Cell (HepG2, HUH-7, and SK-Hep-1 ) was revealed in the 100 ixmol/L Vitamin K2 groups as compared with the control group. Conclusion Vita- min K2 inhibits the proliferation of human hepatocellular carcinoma cell ( HepG2, HUH-7 and SK-Hep-1 ) and Down-regulates the Expression of HDGF.
