摘 要:目的:探讨白藜芦醇以及有氧运动对高脂饮食大鼠血脂代谢、抗氧化及血管内皮功能的影响。方法:40只SD雄性大鼠随机分为正常饮食对照组(N组)、高脂饮食组(H组)、高脂饮食结合运动组(HE组)以及白藜芦醇和高脂饮食结合运动组(HER组),每组10只。运动组大鼠进行3天适应性无负重游泳30 min,以后隔日增加10 min,逐渐延长至90 min,并维持此量,每周训练5天,共8周。白藜芦醇组每天按50 mg/kg剂量灌喂白藜芦醇,其他组灌喂同剂量生理盐水。正常饮食组采用普通饲料,高脂饮食组采用高脂饲料。末次训练结束后,禁食8 h,乙醚麻醉,股动脉取血,肝素抗凝,制备血浆。检测反映血脂代谢的指标总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、载脂蛋白AI(apoAI)、载脂蛋白B100(apoB100)和脂蛋白a[Lp(a)];反映抗氧化能力的指标超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)、丙二醛(MDA)和氧化型低密度脂蛋白(OX-LDL)以及反映血管内皮功能的指标内皮素-1(ET-1)、降钙素基因相关肽(CGRP)和一氧化氮(NO)。结果:与N组比较,H组大鼠TC、LDL-C、OX-LDL、MDA、apoB100、Lp(a)、ET-1显著上升(P<0.05或P<0.01),而HDL-C、apoAI、SOD、CGRP、NO显著下降(P<0.05或P<0.01)。与H组比较,HE组大鼠TC、LDL-C、OX-LDL、MDA、apoB100、Lp(a)、ET-1显著降低(P<0.05或P<0.01),HDL-C、apoAI、SOD、CGRP、NO显著上升(P<0.05或P<0.01)。与HE组比较,HER组大鼠TC、LDL-C、OX-LDL、MDA、Lp(a)、ET-1呈下降趋势(P<0.05),HDL-C、apoAI、SOD、CGRP呈升高趋势(P<0.05)。结论:高脂饮食大鼠血脂紊乱,血管内皮功能下降,抗氧化能力明显减弱。有氧运动减轻高脂饮食大鼠血脂紊乱程度,提高血管内皮功能,增强机体抗氧化能力。有氧运动同时补充白藜芦醇的效果优于单纯运动。Objective To investigate the effect of metabolism, anti-oxidation capacity and the function were randomly divided into four groups:normal diet Resveratrol and aerobic exercise on the serum lipid of vascular endothelial cells. Methods Forty SD rats control group (group N),high fat diet group(groupH) ,high fat diet plus exercise group (group HE) and high fat diet plus exercise plus Resveratrol group (group HER). Rats in exercise groups underwent adaptive 30 min swimming for three days,then the duration increased 10 min every other day till reached 90 min,thereafter the rats continued swimming at this load 5 days per week for 8 weeks. Resveratrol preparation was daily given to rats in group HER with the dose of 50 mg/kg. 8 hours after the last training,total cholesterol (TC),triacylglycerol (TG),high density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), apolipoprotein AI (apoAI), apolipoprotein B 100 (apoB 100), lipoprotein a (Lp (a)), superoxide dismutase (SOD), glutathione peroxidase (GSH-PX), malondialdehyde (MDA), oxidized low-density lipoprotein (OX-LDL), endothelin -1 (ET-1),calcitonin gene-related peptide (CGRP),and nitric oxide (NO) were determined. Results Compared with group N, TC, LDL-C, OX-LDL, MDA, apoB 100, Lp (a)and ET- 1 significantly increased (P 〈 0.05 or P 〈 0.01 ), and HDL-C, apoAI, SOD, CGRP and NO significantly decreased in group H (P 〈 0.05 or P 〈 0.01 ). Compared with group H, the TC, LDL-C, OX-LDL, MDA, apoB 100, Lp (a) and ET-1 decreased significantly (P 〈 0.05 or P 〈 0.01),and HDL-C,apoAI,SOD,CGRP and NO increased significantly (P 〈 0.05 or P 〈 0.01 )in group HE. Compared with group HE ,TC, LDL-C, OX-LDL, MDA, Lp(a) and ET-1 tended to decrease(P 〈 0.05),and HDL-C,apoAI,SOD and CGRP tended to increase (P 〈 0.05) in group HER.
