摘  要:目的:建立RP-HPLC法同时测定黄秋葵不同部位3个黄酮苷[槲皮素-3-O-龙胆二糖苷(HQK-1),槲皮素-3-O-[β-D-木糖基-(1→2)]-α-D-葡萄糖苷(HQK-2),槲皮素-4″-O-甲基-3-O-β-D-葡萄糖苷(HQK-3)]的含量。方法:采用SunFire C18(4.6 mm×250 mm,5.4μm)色谱柱,流动相为甲醇-0.1%磷酸水溶液(47∶53),流速0.8 mL.min-1,检测波长255.6 nm,柱温28℃。结果:本方法可在15 min内完成对HQK-1、HQK-2、HQK-3的色谱分析,且各成分色谱峰之间均达到基线分离;各测定成分的线性范围分别为0.632~9.48μg(r=0.9992),0.065~0.975μg(r=0.9995)和0.102~1.53μg(r=0.9990);平均加样回收率(n=9)分别为100.9%,98.4%,101.8%。以黄秋葵不同部位的3个黄酮苷类成分为研究对象,测定了黄秋葵种子、花、果实、叶、皮中各黄酮苷的含量。结论:结果表明黄秋葵花、种子、果实中黄酮含量较丰富,叶、皮中含量相对较少。本研究所建立的含量测定方法为黄秋葵的入药及资源利用提供了参考依据,可用于黄秋葵中黄酮类成分的定量分析。Objective: To develop an RP - HPLC method for simultaneous determination of 3 glycosylflavones, quer- cetin - 3 - O - gentiobiopyranoside ( HQK - 1 ) , quercetin - 3 - O - [ β - D - xy] - ( 1--.2 ) 1 - β - D - glucopyrano- side ( HQK - 2 ) and quercetin - 4" - O - methy - 3 - O -β - D - glucopyranoside ( HQK - 3 ) in Abelmoschus escu- lentus. Methods:The sample was separated on a SunFire C18column(4.6 mm× 250 mm,5.4 μm) with a mobile phase consisting of methanol - 0. 1% acetic acid (47: 53 ) at the flow rate of 0. 8 mL · min ^- 1 ; UV detection wave- length was 255.6 nm and the column temperature was set 28 ℃. Results:The results showed that all the 3 compo- nents were well separated in 15 rain by RP - HPLC method. The method displayed good linearity of HQK - 1, HQK - 2 and HQK - 3 with the linear ranges and correlation coefficients of 0. 632 - 9.48 μg ( r = 0. 9992 ), 0. 065 - 0. 975 μg( r = 0. 9995 ) and 0. 102 - 1.53 μg ( r = 0. 9990 ), respectively ; The average recoveries ( n = 9 ) were 100.9% ,98.4% and 101.8%. Conclusion:There are fruitful glycosylflavones in A. esculentus and the contents va- ry from flowers to leaves. The HPLC method can be used for the contents determination and resources exploitation of A. esculentus.
