摘  要:目的:探讨自然铜和鹿衔草对肺癌骨转移的抑制作用。方法:取15~20g裸鼠,每只裸鼠胫骨骨髓腔内注射0.1mL A549细胞悬液,14天后建立肺癌骨转移模型。随机分为模型对照组、自然铜组、鹿衔草组和帕米磷酸二钠组。自然铜组和鹿衔草组分别按6g·kg-1隔日自然铜和鹿衔草灌胃,帕米磷酸二钠组按10mg·kg-1每周腹腔注射帕米磷酸二钠,给药3周后处死,称量体重,测量肿瘤体积并计算抑瘤率,流式细胞仪检测肿瘤细胞凋亡率、血生化仪检测血钙、磷、碱性磷酸酶及肝肾功能等。结果:与模型对照组比较,各给药组肿瘤体积均缩小,鹿衔草组和帕米磷酸二钠组细胞凋亡率升高,差异均有统计学意义(P<0.01);各给药组血钙无显著性差异,自然铜组和鹿衔草组血磷升高,帕米磷酸二钠组碱性磷酸酶下降,差异有统计学意义(P<0.01,P<0.05);自然铜组天冬氨酸氨基转移酶升高,差异有统计学意义(P<0.05),帕米磷酸二钠组天冬氨酸氨基转移酶和丙氨酸氨基转移酶升高,尿素氮下降,体重下降,差异有统计学意义(P<0.01)。与帕米磷酸二钠组比较,自然铜组和鹿衔草组抑瘤率、血磷、碱性磷酸酶、体重和尿素氮均升高,丙氨酸氨基转移酶下降,自然铜组细胞凋亡率降低,鹿衔草组细胞凋亡率和天冬氨酸氨基转移酶升高,差异均有统计学意义(P<0.05,P<0.01)。结论:自然铜组和鹿衔草均能抑制肺癌骨转移肿瘤细胞生长,可能机制之一是促进肿瘤细胞凋亡,而且副作用较少。Objective:To discuss the inhibitory action of pyrite and pyrola on bone metastasis in lung cancer. Methods: Take 15 ~ 20g nude mice,inject 0. 1 mL A549 cell suspension into marrow cavity of tibia of every nude mouse, after ld days, establish the models of bone metastasis in lung cancer. Then they were randomLy divided into model control group, pyrite group, pyrola group and pamidronate group. Pyrola group and pamidronate group were separately administered the pyrite and pyrola with 6 g · kg -1 into stomach every other day. Pamidronate group were injected into enterocoelia with pamidronate dis odium 10 mg · kg-1 every week. After three weeks,they were executed and weighed to measure the tumor volume and calcu late the tumor - suppressing rate. Use the flow cytometer to detect apoptosis rate, blood biochemistry analyzer to calcium, phosphorus, alkaline phosphatase,hepatic/renal function and so on. Results:Compared to the model control group,the tumor volume has shrinked in each drug group. Pyrola group and pamidronate group apoptosis rate rised, and there were significant differences (P 〈 0.01 ). Calcium didnt differ significantly among each drug group;phosphorus rised in both pyrite and pyrola group ; alkaline phosphatase declined in pamidronate group ; there were statistical differences (P 〈 0. 01, P 〈 0.05 ). AST went up remarkably in pyrite group(P 〈0.05) ;in pamidronate group,AST and ALT both went up;the urea nitrogen and weight went down;differences were significant(P 〈 0.01 ). Compared to the pamidronate group, anti -tumor rate, phosphorus, al- kaline phosphatase,weight and urea nitrogen all elevated in pyrite and pyrola group;ALT decreased. In pyrite group, apopto- sis rate fell;in pyrola group, apoptosis rate and AST rised. All the differences were significant(P 〈0. 05 ,P 〈0.01 ). Con- clusion: Pyrite and pyrola both can control the growth of cancer cells in bone metastasis caused by lung cancer. One of the possible mechanism is to promote the tumor cell apopto
