石榴皮水煎液对脾虚泄泻模型小鼠血清sIgA等影响的实验研究

摘　　要：目的:观察石榴皮对脾虚泄泻小鼠的止泻作用并初步探讨脾虚泄泻与机体免疫功能之间的关系。方法:以清洁级昆明小鼠40只,随机分组(正常组、模型组、治疗组、对照组),采用苦寒药物灌胃致小鼠脾虚泄泻模型,治疗组以石榴皮水煎液反佐验证治疗,同时设立对照组(思密达),观察实验小鼠一般情况,以放射免疫法测定小鼠血清分泌性免疫球蛋白A(sIgA)含量等指标。结果:造模后小鼠出现活动减少、蜷卧、扎堆、摄食次数减少、大便稀溏等变化,符合脾虚泄泻证表现。治疗组小鼠症状明显改善,血清sIgA含量、T淋巴细胞百分率及转化率、腹腔吞噬细胞百分率及吞噬指数、胸腺/脾脏指数与模型组相比较(P<0.05),治疗组与对照组比较(P>0.05)。结论:石榴皮能改善脾虚泄泻小鼠症状,并能降低造模后小鼠血清sIgA含量,提高T淋巴细胞百分率及转化率、腹腔吞噬细胞百分率及吞噬指数等指标,提示石榴皮的止泻作用可能与调节机体免疫机制有关。Objective：To make an approach to the relationship between the spleen asthenia and immunologic function by observing how granatum invigorating the spleen to arrest diarrhea of the rats of spleen asthenia. Methods：40 clean grade Kunming rats were randomly divided into 4 groups：normal, treated,model,control groups. The model rats were fed with the liquid of folium scnnae to make the spleen asthenia model and treated with the granatum, then their feeding be havior, activity, fur, fecal frequency and so on were observed ; the rats＇ content of slgA, celiac phagocytic ceils percentage and phagocytic index were tested by RIA ;the rats＇thymus and spleen weight change were tested ;the spleen thymus patho- logical changes were tested by HE staining technique. Results ： The model rats appeared to be less active, curling up lying, gathering, feeding rate decreasing,loose stool and so on. It is almost accordant to the symptoms of spleen asthenia. The content of sIgA, celiac phagocytic ceils percentage and phagocytic index in mode group were increased compared with those in the normal one（ P 〈 0.05） . The treated group had obvious improvement in symptoms. The content of sIgA, celi ac phagocytic cells percentage, phagocytic index and immune organs weight in treated group had significance compared with those in the model group （ P 〈 0.05 ）, there was no obvious difference between the treated group and control sample group（P 〉0.05）. Conclusion：Granatum can improve the symptoms of spleen asthenia,it may be related to the gastroin testinal mucosa protection and the immune mechanism.
