摘  要:目的:分析同卵双生子间的差异表达基因,初步探讨双生子2型糖尿病(Type 2 Diabetes Mellitus,T2DM)罹患差异性的可能分子生物学特征。方法:根据双生子卵性相似诊断法,评判双生子的指纹、中指毛、耳垂、毛发等26项指标的相似度,判断双生子的卵性(同卵或异卵)。应用基因芯片技术,以双生子的同胞弟弟为健康对照,筛选同卵双生子(Monozygotic Twins,MZT)之间的差异基因表达谱,并通过FatiGO数据库进行基因功能比较。结果:这对双生子为MZT。与健康对照比较,得到双生子的差异表达基因,大双为968个,小双为478个,二者之间相同的显著表达基因有15个,基因功能比较结果见免疫反应功能类基因及细胞因子通路显著表达。结论:同卵双生子T2DM患病的差异性主要与个体的免疫功能差异有关。Objective:To analyze significant expressed genes of MZ twins and analyze molecular biological foundation of variability between T2DM twins. Methods :According to the similarity diagnosis of twins zygosity,the resemblance of 26 items such as fingerprint,the hair on middle finger, ear lobe,hair and so on were assessed, and the zygosity( MZ or DZ)of twins was further determined. The significantly expressed genes between each of twins and healthy control were screened through cDNA microarray test. These screened genes were further functionally annotated through FatiGO database. Results : The twins were MZ twins. 968 and 478 significantly expressed genes were screened for the older and younger sister respectively. Fifteen identical genes were selected from significantly expressed genes of twin sisters. GO and PATHWAY annotation results of 15 genes showed that immune-related genes and Cytokine-eytokine receptor interaction pathways significantly expressed. Conelusion:The dissimilarity of T2DM between the MZ twin sisters is mainly related to differences in the individualg immune function.
