摘  要:分别采用冰水混合物、0.7 mmol/L环己酰胺和0.2%秋水仙素3种试剂,在多个时间梯度对生长旺盛的杂种鹅掌楸体胚苗根尖进行预处理,并采用压片法进行制片观察。结果表明:采用冰水混合物处理6 h,0.7 mmol/L环己酰胺处理2 h,0.2%秋水仙素处理4 h,均可获得较多的根尖细胞中期分裂相。相比较而言,采用环己酰胺和秋水仙素作为预处理试剂获得的染色体形态较好。利用0.7 mmol/L环己酰胺对鹅掌楸根尖染色体进行2 h的预处理后,采用卡宝品红压片法制片进行染色体核型分析的结果显示,杂种鹅掌楸染色体为38条,核型公式为2n=38=26m+8sm+2st+2t,核型属于2B型。Ice-water mixture,0.7 mmol/L cycloheximide and 0.2 % colchicine were used as pretreated reagent for Liriodendron hybrids root tips of test-tube plantlet by different time grads.Then the root tips were squashed with Carbol fuchsin and observed with microscope.It had been proved that all these three pretreatments of ice-water mixture for 6 h,0.7 mmol/L cycloheximide for 2 h,and 0.2 % colchicine for 4 h could obtain metaphase chromosome specimen,but by comparison,root tips pretreated with cycloheximide and colchicine could give much better chromosome morphology.Then Karyotype of the Liriodendron chinense×L.tulipifera was studied with chromosome samples prepared from root tip by 0.7 mmol/L cycloheximide for 2 h.The result showed that there were 38 chromosome in Liriodendron chinense×L.tulipifera and its karyotype formula was 2n=38=26m+8sm+2st+2t and the karyotypes of Liriodendron chinense×L.tulipifera belong to category 2B of Stebbins.
