摘  要:目的探讨神经管发育缺陷(NTD)发生过程中与正常组织差异表达的相关基因,并作初步功能分类。方法用含有1100余个已知基因的中密度芯片比较了胚胎9.5d、10.5d正常与同期NTD小鼠神经管组织的基因表达差异。结果通过比较正常E9.5d与E10.5d获得138个差异表达基因,E9.5d-NTD获得20个差异,E10.5d-NTD获得29个差异表达基因,根据基因功能分类包括细胞周期与凋亡相关基因,信号转导基因,转录与翻译调控,能量与代谢基因,热休克基因,基质与骨架蛋白等多种种类。结论实验证实多类基因参与了NTD的发生、发展过程,对相关基因群的进一步研究有助于阐释NTD的发病机制。Objective To investigate genes in neural tube defects(NTD)different from normal tissue,and to take a preliminary functional classification.Methods To compare the difference of gene expression in the normal embryo and the same period of NTD neural tube organizations within 9.5 days and 10.5 days using medium density chip which contains more than 1,100 known genes.Results There were 138 different genes in the normal embryo on E9.5d contrast on E10.5d.There were 20 different genes in NTD on E9.5d,and 29 different genes on E10.5d.A classification based on gene function included genes related to cell cycle and apoptosis,signal transduction genes,transcribel and translational regulation genes,energy metabolism genes,heat shock genes,matrix and cytoskeletal proteins genes etc.Conclusion The experiment presents multiclass genes involved in the process of the occurrence of NTD,so further study on the related genes would help to explain the pathogenesis of NTD.
