摘  要:为探讨γ射线对丝素蛋白生物相容性的影响,用不同剂量(0、25、50、100和200kGy)γ射线对丝素蛋白进行辐照,通过检测丝素蛋白结构变化、细胞增殖活力、细胞毒性以及溶血性检验,探索不同剂量γ射线处理对丝素蛋白生物相容性的影响。结果表明:在本实验所设定的剂量范围内,γ射线辐照对丝素蛋白结构影响不明显,照射后丝素蛋白对细胞增殖活力的影响较小,浸提液对细胞无明显毒性,丝素蛋白材料及其浸提液的血液相容性良好。结论是,在0-200 kGy剂量范围内,γ射线辐照对丝素蛋白结构及其生物相容性影响均不明显。To investigate the effects of γ-irradiation on the biocompatibility of silk fibroin, the silk fibroin film was irradiated with different absorbed doses of 0, 25, 50, 100 and 200 kGy, respectively. The dermal cells were used as experimental cell model and cultured on the irradiated films. The molecular structure of silk fibroin was analyzed by FT-IR (Fourier Transform infrared spectroscopy). The graded toxicity and hemolysis had been tested. The results showed that there was no obvious change with the molecular structure of silk fibroin in the range of absorbed dose, meanwhile there was no significant effects on the growth and proliferation activity of dermal cells. And there was also no obvious toxicity to the cells and the silk fibroin film had the good hemo-compatibility. In conclusion, in the range of 0- 200 kGy, the γ-irradiation has no significant effects on the structure and biocompatibility of silk fibroin.
