摘 要:构建了含有Bt杀虫基因cry1Ah和耐草甘膦基因2mG2-epsps的双价植物表达载体pMAGUHM,通过基因枪轰击Q31×Z31杂交组合的胚性愈伤组织,以2mG2-epsps为筛选标记,通过1mmol/L草甘膦异丙胺盐筛选,获得T0代转化植株80株,其中PCR检测有36株为阳性植株,阳性率达45%。对得到的阳性植株的T1、T2代进行了跟踪检测,证明外源基因已整合到玉米基因组中并正确表达。草甘膦及玉米螟抗性检测结果表明,有5个转基因品系表现出具有耐草甘膦和抗虫的双重特性。A bivalent plant expression vector pMAGUHM expressing the modified Bt crylAb gene and glyphosate- tolerant 2mG2-epsps gene was constructed and transformed into Zea mays L. calli Q31 × Z31 hybrid by microprejec- tile bombardment. Using the 2mG2-epsps gene as a selective marker, eighty glyphosate-tolerant maize plantlets were selected by l mmol/L glyphosate- isopropylammonium, and thirty-six glyphosate-tolerant maize plantlets were con- firmed transgenic plants by PCR analysis. The positive rate was 45%. Further analysis was performed on the T1 and T2 generation and it was confirmed that Bt crylAh gene and 2mG2-epsps gene had been integrated into the maize genome and expressed successfully. The bioassay of corn borer (Pyrausta nubilalis Hubern) resistance and glypho- sate tolerance indicated that five transgenic lines showed both glyphosate tolerant and insect resistant traits.
