盲肠结扎穿孔脓毒症大鼠血清DAO与CGRP变化及其相关性研究

摘　　要：目的通过观察盲肠结扎穿孔脓毒症大鼠血清DAO、CGRP水平变化及相关性分析,探讨CGRP在肠黏膜损害中的作用。方法将60只SD大鼠随机分成脓毒症组和假手术组,每组30只,脓毒症组采用CLP制作大鼠脓毒症模型,假手术组仅行盲肠探查术。各组均于术后3、6、12、24与48 h时点,采集腹主动脉血通过双抗体夹心酶标免疫分析法检测血清DAO、CGRP水平,并做相关性分析。结果脓毒症组中的DAO水平与假手术组比较,CLP术后3 h明显升高(P<0.05),呈持续上升趋势,在CLP术后48 h血清DAO水平明显高于假手术组(P<0.05)。脓毒症组中的CGRP水平与假手术组比较,CLP术后3 h明显降低(P<0.05)呈持续降低趋势,虽随时间推移有所回升,但在CLP术后48 h仍低于假手术组(P<0.05)。血浆CGRP、DAO含量的变化,经相关分析,CGRP与DAO具有高度正相关,相关系数为0.904,(P<0.01)。结论盲肠结扎穿孔脓毒症大鼠DAO明显高于正常水平、CGRP呈现持续降低状态,CGRP的释放减少可能是导致肠缺血缺氧和肠黏膜损伤的因素之一。Objective To investigate the role of（calcitonin gene-related peptide,CGRP）and（diamine oxidase,DAO） in intestinal mucosa damage of septic rats.Methods Sixty SD rats were randomly separated into septic group and sham operation group（n=30 in each group）.Septic rats were obtained by CLP surgery.Only the exploratory cecum surgery was conducted in rats of sham operation group.3 h,6 h,12 h,24 h,48 h after surgery,ventral aorta blood was collected for ELISA detection of CGRP and DAO.Results DAO level of septic rats was dramatically increased at 3 h（P〈0.05） after the CLP surgery and continually increased in the subsequent 48 h.CGRP level of septic group was significantly decreased 3 h after the CLP surgery（P〈0.05）.Although the CGRP level was generally increasing within 3-48 h after CLP surgery,the CGRP level of septic group was still significantly lower than that of the sham group（P〈0.05）.Through the correlation analysis,DAO and CGRP showed a high positive correlation（r=0.904,P〈0.01）.Conclusion Acute damage of intestinal damage by sepsis will induce continual decrease of CGRP and increase of DAO in blood.The study indicates the short of CGRP release might be an important factor of sepsis induced intestinal ischemia,anoxia and acute intestinal damage.
