摘  要:为查明在广州罗非鱼良种场获得的第一批尼罗罗非鱼♀(Oreochromis niloticus)×萨罗罗非鱼♂(Sarotherodonmelanotheron)杂交F1群体的亲本信息,以原杂交试验中的亲本群体及杂交F1为对象,利用微卫星标记和基因重组法推测F1的亲本数目。结果发现:86对微卫星引物中有74对稳定扩增;筛选出14对在尼罗罗非鱼♀和萨罗罗非鱼♂中存在不同等位基因的引物,对杂交F1进行扩增,发现尼罗罗非鱼♀与萨罗罗非鱼♂的等位基因在F1中均结合为杂合基因型,证实杂交F1为真杂交种;应用基因型重组法根据各位点在F1中的基因型,对比其亲本群体的特异等位基因,推测F1中各位点的等位基因是来自于母本或父本,进而判断F1真实有效的父母个体的等位基因,推测出最小的有效亲本数目为一雌一雄,表明F1可能互为全同胞,为尼萨罗非鱼杂交种的进一步选育提供了基础资料。In order to investigate the parental information of the first Oreochromis niloticus ♀xSarotherodon melanotheron ♂ hybrid F1 obtained at National Tilapia Seed Farm of Guangdong, the effective parental number were analysis by microsatellites and genotypic reconstruction with the hybrid F1 and its parent populations. The results showed that 74 microsatellite primers were amplified stably. 14 primers were used to amplify Ft selected with different size alleles in O. niloticus ♀and S. melcotheron ♂, and it found that F1 carried one male parent and one femal parent molecular marker at each polymorphic locus and F1 was identifyed the genuineness of hybrid from Oreochromis niloticus ♀ xSarotherodon melanotheron♂. Whether these F1 alleles at each polymorphic locus coming from male parent or female parent was analyzed using genotypic reconstruction comparing those specific alleles in its parent populations, and then alleles of really effective parents were inferred, the minimum of effect parents number was conjectured to be one male and one female which showed the F1 might be full-sibs, which provided data for the selective breeding of new salt-tolerant tilapia.
