摘  要:采用显微镜计数法、荧光分光光度法、叶绿素a含量测定法、可见分光光度法对利玛原甲藻(Prorocentrum lima)指数生长期不同浓度的培养液进行细胞密度测定。结果表明,后3种细胞计数方法与显微镜计数法相比较,均有较好的线性关系;同时对不同生长期的利玛原甲藻培养液进行测定,结果表明,4种方法的生长曲线一致性良好。比较分析发现,可见分光光度法适用于不同细胞密度范围的测定,操作简单、耗时短、不需要破坏样品、适于批量操作,可确定为便捷、有效、可行的利玛原甲藻细胞计数法。In this study, four cell-counting methods of microscope, Fluorescence Spectrophotography, Visible Spectrophotography and Chla Containing Measurement were used to measure the amount of the exponential phase cell of Prorocentrum lima. The insightful comparison on the interrelations of the last three mentioned cell counting methods was made with the method of microscope, the results revealed the sound linear interrelations among them. Different growth phase of P. lima were simultaneously measured, and then growth curves were drew basis on the data from different methods. The results showed good concordance between these measurement methods. Particularly, for Visible Spectrophotography Methods, the operating procedures were simple, fast, suitable for wide cell-density ranges, batch measurements and the test sample ceils were not destructed and could be reused for further research after measurement. Therefore, visible spectrophotography was convenient, high-efficiency and reliable approach for cell counting of P. lima.
