摘  要:以胡萝卜‘长城红玥’与‘开拓者’胚轴为受体材料,研究了菌种、预处理、农杆菌菌液浓度、侵染时间和共培养时间对胡萝卜胚轴转化率的影响。结果表明:C58C1菌株对胡萝卜胚轴伤害较小;‘长城红玥’以7日龄无菌苗于4℃预处理3d,经3.2×107个/mL农杆菌菌液侵染6min,共培养3d,转化率可达60%;‘开拓者’以7日龄无菌苗于4℃预处理3d,经4.8×107个/mL农杆菌菌液侵染10min,共培养4d,或7日龄无菌苗于4℃预处理10d,经3.2×107个/mL农杆菌菌液侵染10min,共培养2d转化率可达100%;但2个品种均以7日龄无菌苗于4℃预处理3d,经1.6×107个/mL农杆菌菌液侵染2min,共培养2d,可得最多转化苗。Taking the hypocotyls of 'Changchenghongyue' and 'Kaituozhe' of Daucus carota L. as materials,the effect of the strains, the condition of pretreatment, concentration of Agrot)acteriurn suspension, the time of infection and the time of co-cultivation on the transformation of carrot 'Changchenghongyue ' and 'Kaituozhe ' hypocotyls were studied. The results showed that the C58C1 strain was less damaged for carrot hypocotyls;and the transformation frequency was up to 60% for hypocotyls of 'Changchenghongyue' ,which derived from pretreatment 7-day-old aseptic plantlets at 4℃ for 10 days, infected with suspension of A. tumefactions at the concentration of 3. 2 × 10^7 cells/mL for 6 rains,co-cultivated for 3 days;and the transformation frequency was up to 100% for hypocotyls of 'Kaituozhe' ,which derived from pretreatment 7-day-old aseptic plantlets at 4℃ for 3 days,infected with suspension of A. turnefactions at the concentration of 4. 8 × 10^7 cells/mL for 10 min,co-cuhivated for 4 days or derived from pretreatment 7-day-old aseptic plantlets at 4℃ for 10 days, infected with suspension of A, tumefactions at the concentration of 3. 2 ×10^7 cells/mL for 10 min, co-cultivated for 2 days;and the transformants were more for both of their hypoc0tyls, which derived from pretreatment 7-day-old aseptic plantlets at 4℃ for 3 days,infected with suspension of A. tumefactions at the concentration of 1.6 × 10^7 cells/mI, for 2 mins,co-cultivated for 2 days.
