膀胱移行细胞癌多肿瘤抑制基因-1纯合子缺失及意义

摘　　要：目的 :了解多肿瘤抑制基因 - 1(MTS1 )在膀胱移行细胞癌中的纯合子缺失情况 ,探讨 MTS1 纯合子缺失在膀胱移行细胞癌发生与发展中的意义。方法 :收集温州医学院手术切除的膀胱移行细胞癌石蜡标本 35例 ,采用比较 PCR法检测其 MTS1 纯合子缺失情况。结果 :剔除 2例不可比标本 ,33例标本中 ,MTS1 纯合子缺失 6例 ,缺失率为 18.2 %。其中 级、 级、 级膀胱移行细胞癌 MTS1 纯合子缺失率分别为 18.2 % (2 / 11)、2 0 .0 % (3/ 15 )和 14.3% (1/ 7) ;Tis～ T1 期肿瘤和 T2 ～ T4期肿瘤 MTS1 纯合子缺失率分别为 2 2 .2 % (4 / 18)和 13.3% (2 / 15 ) ,各级和各期肿瘤的 MTS1 纯合子缺失率无显著性差异 (P >0 .0 5 )。结论 :MTS1 纯合子对膀胱移行细胞癌的发生起一定作用 。Purpose:To understand the states of homozygous deletions of the multiple tumor suppressor gene 1(MTS1) in the transitional cell carcinoma of the bladder and their significance. Methods:Paraffin embebed specimens from 35 cases of patients with transitional cell carcinoma of the bladder was detected by the comparative PCR for homozygous deletions of MTS1.Results:Discarding 2 samples which was not comparable,homozygous loss of MTS1 was found in 6 of 33 samples ,loss rate was 18.3 %.The loss rate in grade Ⅰ,Ⅱ and Ⅲ tumor was 18.2 % (2/11), 20.0 % (3/15) and 14.3 %(1/7) respectively;the loss rate in the superficial tumors (Tis,T1) and the invasive tumors was 22.2 % (4/18) and 13.3 % (2/15) respectively.There wasn′t statistically significant difference between histological grades and between pathological stages( P > 0.05 ).Conclusions:The results showed that the homozygous deletions of MTS1 was correlated with the development in some transitional cell carcinoma of the bladder, but wasn′t correlated with the progression.
