摘  要:目的研究酸性丝氨酸蛋白酶ASPNJ单独用药及与多柔比星联合用药对体外培养的人慢性髓系白血病细胞株K562的抑制作用,并初步探讨其作用机制。方法应用MTT比色法、台盼蓝拒染法检测细胞增殖和细胞活力,倒置显微镜下观察细胞的动态变化,瑞氏-吉姆萨染色法观察细胞的形态改变,流式细胞术检测细胞凋亡。结果沙蚕丝氨酸蛋白酶ASPNJ有效抑制K562细胞的增殖和活力并呈浓度和时间依赖性,联合用药显示ASPNJ显著增加多柔比星的杀伤作用;K562细胞出现凋亡所具有的形态学变化,ASPNJ明显诱导K562细胞的凋亡。结论沙蚕丝氨酸蛋白酶ASPNJ对体外培养的K562细胞具有明显的抑制作用并能增强多柔比星的效应,其作用机制可能与直接的细胞毒性作用及诱导凋亡有关。Purpose To investigate the effects of Acidic Serine Protease ASPNJ on inhibition of chron- ic myeloid leukemia cell lines K562, cells were treated with different concentrations of ASPNJ alone and combined with application of Doxoruhicin. Methods MTT assay and trypan blue exclusion test were used to observe the inhibitory effect. The growth of cells was observed continuously under the inverted micro- scope. The morphological changes of cells were examined by Wright's-Giemsa staining. The effect of apop- tosis on cells was determined by flow eytometry. Results The proliferation and viability of K562 cells were inhibited definitely by ASPNj in a dose-and time-dependent manner, and ASPNj also enhanced the effect of Doxoruhicin. K562 cells showed the morphological changes and increased apoptosis by ASPNj. Conclusion ASPNj strongly inhibits proliferation and viability of K562 cells in vitro and enhances the effectiveness of Doxoruhicin. The mechanism may be related with the direct cytotoxic effect and induction of apoptosis.
