经后增殖方和促黄体方能诱导超促排卵小鼠子宫内膜中整合素β_3的表达

摘　　要：目的:观察益气血补肝肾方对控制性超促排卵小鼠子宫内膜组织中整合素β3表达的影响。方法:将实验小鼠随机分为COH模型组、中药治疗组和对照组。进入实验后,中药治疗组每日给予中药灌胃,其余小鼠以等量生理盐水代替。COH模型组和中药治疗组给予GnRHa降调节,PMSG+hCG促排。每组分别于孕第1日(pd1)、pd2、pd4、pd5、pd6处死10只动物,取子宫内膜采用实时荧光定量RT-PCR和免疫印迹的方法分别在mRNA和蛋白水平定量检测子宫内膜整合素β3表达情况。于妊娠第8日(pd 8)剖腹取各组小鼠子宫(每组10只),观察胚泡着床情况。结果:整合素β3在各组小鼠子宫内膜中呈周期性的表达,从孕第1日(pd 1)开始整合素β3表达逐渐增强,pd 4显著增加,pd 6表达开始减弱。与COH模型组相比,中药治疗组小鼠子宫内膜中整合素β3表达增强,两者相比有统计学意义(P<0.05);整合素β3在中药治疗组和对照组中的表达水平相近,两者相比无统计学意义(P>0.05)。COH模型组pd 8的平均胚胎着床数与中药治疗组和对照组相比,明显减少(P<0.05),而中药治疗组与对照组相比无统计学差异(P>0.05)。结论:益气血补肝肾方可诱导小鼠子宫内膜中整合素β3的表达,并提高超促排卵小鼠的胚胎着床率,提示该方可能具有改善子宫内膜容受性的作用。To observe the effect of Yiqixuebuganshen recipe on uterine expression level of integrin β3in the controlled ovarian hyperstimulation （COH） mice. Methods： A total of 180 mice were randomly divided into 3 groups： untreated COH group, treated COH group and control group. Real-time RT-PCR and Western blotting were used to detect the uterine expression levels of integrin β3 mRNA and integrin β3 protein, respectively. The mean number of implanted embryos of the three groups was calculated on day 8 of pregnancy （pd 8）. Results： Integrin β3 expressed regularly in the endometrium of COH mice. Its expression level increased gradually along with pregnancy progressing. Integfin β3 expression level significantly increased on pd 4, and decreased on pd 6. Compared to the untreated COH group, the expression level ofintegrin β3 in the treated COH group was remarkably up-regulated, and the difference was significant （P〈0.05）. Whereas the difference between the treated COH group and the control group was not significant （P〉0.05）. Furthermore, the average number of implanted embryos of the untreated COH group was significantly less than the other two groups （P〈0.05）; Whereas the difference between the treated COH group and the control group was no significance （P〉0.05）. Conclusion： Yiqixuebuganshen recipe could induce the uterine expression of integrin β3 in mice, indicating it might improve the endometrial receptivity and promote the embryo implantation.
