摘 要:目的探讨神经酰胺与颗粒细胞凋亡的关系。方法 以PMSG处理的雌性Wistar大鼠为模型,制备了颗粒细胞悬液。应用TUNEL法及流式细胞术检测了神经酰胺对离体培养颗粒细胞凋亡的影响。 结果 体外培养的颗粒细胞存在自发性凋亡,神经酰胺处理的颗粒细胞凋亡率明显高于对照组(P<0.05);流式细胞术分析可见增强的凋亡峰。结论神经酰胺可能作为一种重要的信息分子参与大鼠颗粒细胞凋亡。Objective To investigate the relationship between ceramide and apoptosis of granulosa cells. Methods PMSG-treated female Wistar rats were used as a model and granulosa cell suspensions were pared. The effect of ceramide on apoptosis of granulosa cells was assessed by TUNEL method and flow cytometry analyses. Results There spontaneous apoptosis in the cultured granulosa cells and ceramide could significantly increase apoptosis (P<0.05). Enhanced apoptotic surf was observed by flow cytometry. Conclusion Ceramide may serve as an important signaling molecule, mediating rat granulosa cell apoptosis.
