摘 要:为获得高诱导率的花生毛状根诱导条件,同时探究不同浓度的萘乙酸(NAA)和2,4二氯苯氧乙酸(2,4-D)对花生毛状根生长和白藜芦醇含量的影响。研究了不同发根农杆菌菌株和不同乙酰丁香酮浓度对花生毛状根诱导率的影响;以固体MS培养基为对照,分别添加不同浓度的NAA和2,4-D,培养花生毛状根,探究对毛状根生长和白藜芦醇含量的影响。实验显示花生毛状根诱导率可达到92.3%。花生毛状根随着NAA的浓度升高,生物量同对照相比较无显著差异,各浓度的NAA之间也无明显差异;与对照相比较,白藜芦醇含量显著增高,呈显著差异,但各浓度的NAA之间白藜芦醇的含量无明显差异。添加2,4-D形成的生物量明显偏低,白藜芦醇含量与添加NAA组无差别。以A600为0.8的发根农杆菌R1601菌液侵染花生叶片,在含50μmol/L乙酰丁香酮的MS培养基培养时,可以高效的诱导得到花生毛状根;NAA可以作为一种外源激素提高花生毛状根中白藜芦醇的含量。To obtain induction conditions about a high induction rate of peanut hairy root, and to explore the influence of biomass and resveratrol content of peanut hairy root with different concentrations of naphthalene acetic acid (NAA) and 2,4-phenoxyacetic acid (2,4-D) , the effects of induced rate of peanut hairy roots with different strains of Agrobacterium rhizogenes and different aceto- syringone concentrations were studied and Ms solid medium was used as control, then different concentrations of NAA and 2,4-D were added to cultivate peanut hairy roots, and to explore the influence of hairy root biomass and resveratrol content. The results showed that the induction rate of peanut hairy root reached 92.3 %. With increasing NAA concentration, no significant difference was observed in the biomass accumulation between the control and the group treated with various concentrations of NAA. With NAA concentration more and more higher, compared with control, resveratrol content was significantly increased, showing a significant difference, but the resveratrol content had no significant difference in the different concentrations of NAA. Adding 2,4-D, the biomass is significantly lower, the resveratrol content had no difference compared with NAA group. The leaves of peanut were infected by Agrobacterium rhizogenes R1601 ( A600 = 0.8 ) , and cultivated the infected leaves on MS solid medium that contained 50 μmol/L acetosyringone, the hairy roots of peanut can be efficiently induced and NAA can serve as an external hormones to increase the resveratrol content in hairy roots of peanut.
