摘 要:目的观察存活素(Survivin)基因转染对大鼠骨髓间充质干细胞的体外抗凋亡作用。方法 PCR扩增Survivin全长编码基因,通过脂质体介导将Survivin基因转染至骨髓间充质干细胞,RT-PCR检测转染后Survivin mRNA的表达,Annexin V/PI双标法检测去血清诱导后细胞凋亡比例。结果 RT-PCR显示转染组有效表达Survivin mRNA,转染组去血清培养后的早期凋亡率较未转染组降低,差异有统计学意义(P<0.05)。结论 Survivin基因转染对骨髓间充质干细胞有保护性作用,可增强其抗凋亡能力,延长其存活时间。Objective To observe the in vitro biological characteristics and anti-apoptotic effects of rat bone mesenchymal stem ceils (BMSCs) transfected with Survivin. Methods The Survivin coding sequence was amplified by polymerase chain reaction (PCR) method and sub- cloned into liposome. The plasmid was transfected into BMSCs, and RT-PCR method was used to detect the Sunvin expression. Annexin V- PI double staining method was used to detect cell apoptosis proportion. Results RT-PCR results confirmed that the transfected group effec- tively express Surcivin mRNA compared with controls. Early apoptosis rate of the transfected group was significantly lower than the control group (P 〈 0.05 ). Conclusion Survivin gene transfection can enhance the anti-apoptotic ability of the BMSCs so as to extend their sur- vival time.
