血清脂联素水平及其基因276位点多态性与动脉粥样硬化的关系

摘　　要：目的:探讨在台州地区动脉粥样硬化人群中脂联素血清水平及其基因SNP+276多态性与动脉粥样硬化之间的关系。方法:收集2009年7月-2011年12月进行过冠状动脉造影住院患者共512例,根据冠脉造影结果狭窄>50%为病变组(AS)和<50%为对照组,用Gensini评分系统评价冠状动脉损害严重程度,采用PCR-RFLP方法,分析其基因276位点的多态;采用ELISA法测定血清脂联素水平。结果:AS组血清脂联素水平低于对照组。且其276位点TT基因型和T等位基因频率均低于对照组;Logistic多因素回归分析显示:GG+GT型个体患AS的相对危险度是TT型的2.324倍。结论:在台州地区人群中,血清脂联素水平与AS呈负相关,基因SNP+276 G/T与AS的发生相关,携带GG+GT型者患AS的危险因素增加,G等位基因是AS的危险因素。Objective：To understand the role of ＋276 polymorphisms and serum expression level of adiponectin in atherosclerotic（AS） population of Taizhou.Methods： A total of 512 atherosclerotic patients with the confirmation by Coronary angiography were included from July 2009 to December 2011.According to angiography,the artery stenosis 50% belong to AS group,50% belong to control.The severity of coronary artery stenosis was evaluated and scored with Gensini coronary scoring system.Adiponectin polymorphisms were tested by PCR-RFLP;serum adiponectin level were detected by ELISA.Results： AS group had a low level of serum adiponectin and low frequency of TT and T allele at 276 site when compared with control group.Further logistic regression analysis reveled that GG＋GT allele had a 2.324 folder higher risk in the atherosclerotic occurrence.Conclusion： In Taizhou population,the serum expression level and 276G polymorphism of adiponectin gene are risk factors of AS occurrence.
