不同剂量丙酸睾酮对半去势小鼠生精功能的影响

摘　　要：目的探讨不同剂量丙酸睾酮对半去势小鼠生殖系统的发育及精子发生的影响。方法将50只性成熟的昆明小白鼠随机分成5组（每组10只），A组为假手术组，B、C、D、E4组为半去势组（切除右睾丸）。半去势组组中除E组为半去势空白组（术后每天注射生理盐水12mg／kg）外，其余3组按术后每天肌注两酸睾酮剂量1．5mg／kg、3mg／kg和12mg／kg分为半去势小剂量T组（B组）、半去势中剂量T组（C组）和半去势大剂量T组（D组）。一周后，检测各组小鼠外周血雄激素水平，精子活力、活率、畸形率，生殖器重量指数等指标。结果精子活力在半去势对照组显著低于假手术组（P〈0．01），中剂量丙酸睾酮组显著高于假手术组（P〈0．01）；精子畸形率则半去势各组均显著高于假手术组（P〈0．05）；精子存活率在中剂量睾酮组显著高于半去势对照组（P〈0．05）。手术对侧睾丸指数随用药量的增加而增高，达到12mg／kg时，睾丸指数最高为（3．30±0．22），显著高于半去势对照组（P〈0．05）。手术同侧附睾指数半去势后均减小，补充睾酮后有所增加，但差异无统计学意义（P〉0．05）。结论适量丙酸睾酮能提高半去势后精子总活力、存活率，降低精子畸形率；对侧睾丸随肌注丙酸睾酮剂量的增加而增大。Objective To investigate the effect of different doses of testosterone propionate on the reproductive organ performance and spermatogenesis in semi-castrated mice. Methods Ten healthy mice selected from 50 male Kunming mice with mature sexual function were used d as the sham operation group. The remaining 40 mice under- went removal of their right testes and then, they were randomly divided into 4 groups, receiving intramuscularly injections with different doses （1.5mg/kg, 3mg/kg, 12mg/kg） of testosterone propionate and normal saline every day for seven days, Afterwards, the sex hormone levels in peripheral blood and sperm motility, survival rate, aberration of sperm were detected. At the same time, the weight of the testis, epididymis, seminal vesicle and kidney were measured. Results Sperm motility in the semi-castrated group was significantly lower than that in the sham operation group （P〈0.01）. Sperm motility in moderate dose testosterone propionate group was higher than that in sham operation group （P〈0.01）. The sperm deformity rate in the semi-castrated group was significantly higher than that in the sham operation group （P〈0.05）. The sperm survival rate in the group receiving the moderate dose of testosterone propionate was significantly higher than that in the semi-castrated control group （P〈0.05）. The testicle index of the other side after surgery reached the highest （3.30±0.22） by giving 12mg/kg testosterone propionate and it was significantly higher than that in the semi-castrated control group （P〈0.05）. The epididymis index of the surgecal side was reduced the in semi-castrated group, but after supplement of testosterone propionate, it was increased, however, there was no significant difference （P〉0.05）. Conclusion Moderate dose of testosterone propionate can improve the sperm motility, survival rate and reduce deformity rate in the semi-castrated mice. The testicle index of the other side after surgery elevates along side the increase of the dose of testos
