摘  要:目的在建立版纳微型猪糖尿病胰岛素抵抗的模型基础上,观察其肾脏病变及GSK-3β、cyclin D1的表达变化,探讨二者之间可能存在的作用关系。方法高糖高脂饮食加低剂量STZ诱导版纳微型猪糖尿病模型,观察其血糖、胰岛素、肾脏组织结构病理变化,Realtime-PCR检测肾脏组织GSK-3β、cyclin D1 mRNA含量变化,研究二者相关性,探讨其作用机制。结果喂养12个月后,实验组较之对照组出现空腹血糖明显升高(P<0.01),阶段性低胰岛素血症,肾小管上皮细胞水肿,间质胶原纤维增生,部分肾小球囊腔增大,内部细胞数目增加,肾小球肥大,基底膜增厚,GSK-3β、cyclin D1mRNA肾脏组织明显上调。结论高糖高脂饮食加低剂量注射STZ构建了版纳微型猪糖尿病胰岛素抵抗模型,上调肾脏组织GSK-3β、cyclin D1表达,二者表达正相关。Objective On basis of the establishment of the Banna Minipig model of type 2 diabetes insulin resistance, we observed GSK-3β, cyclin D1 expression changes and explored their possible relationship with the pathological changes in the kidney. Methods Banna miniature pigs fed with high fat and high sucrose diet (HFSD) plus low-dose STZ injection caused diabetes mellitus. To observe their blood glucose, insulin and pathological changes of kidney tissue structure, kidney tissue GSK-3β, cyclin DlmRNA content changes were detected by Reahime-PCR. Results Fed with HFSD for 12 months, plasma glucose levels were significantly higher than in the control group (P 〈0.01 ), and we alsoobserved stage hypoinsulinemia, renal tubular epithelial cell edema between prolife ration of collagen fibers, increased number of internal cell, glomerular hypertrophy, basement membrane thickening, and GSK-3β, cyclin DlmRNA in the kidney were significantly increased. Conclusion HFSD diet supplement with low-dose STZ injection more times established Banna miniature pig type 2 diabetes insulin resistance, increased the expressions of GSK-3β, cyclin D1 in the kidney, and both of them showed positive correlation.
