摘  要:目的:观察柴黄益肾颗粒对糖尿病肾病并发肝损伤大鼠的肝保护作用,并探讨其可能机制。方法:单侧肾切除合并链脲佐菌素注射诱导糖尿病肾病大鼠模型,给药20周后,检测大鼠血糖、肝功能、肝组织病理、肝组织Ⅰ、Ⅲ型胶原及肝糖原蓄积程度,并检测肝组织葡萄糖转运蛋白-1(GLUT-1)、转化生长因子-β1(TGF-β1)mRNA及蛋白表达水平。结果:与假手术对照组比较,模型组大鼠肝组织表现为肝细胞索排列紊乱,炎性细胞浸润,糖原颗粒蓄积,Ⅰ、Ⅲ型胶原纤维异常增生,柴黄益肾颗粒组大鼠肝组织炎性细胞浸润、糖原蓄积程度及Ⅰ、Ⅲ型胶原纤维增生程度显著减轻,肝组织GLUT-1及TGF-β1 mRNA、TGF-β1蛋白表达显著低于模型组(P<0.05)。结论:柴黄益肾颗粒对糖尿病肾病并发肝损伤大鼠具有肝保护作用,这可能与柴黄益肾颗粒显著降低肝组织GLUT-1及TGF-β1表达有关。Objective:To investigate the effect of Chaihuangyishen granule (CHYS)the expressions of glucose transporter-1 (GLUT-1)and transforming growth factor β1 (TGF-β1)in the liver of diabetic nephropathy rats, and probe the mechanism.Methods:Male Wistar rats were randomly divided into 12 rats in sham control group and 100 rats in uninephrectomy groups which were induced into diabetic model by injection of 40mg/kg STZ.The diabetic rats were then randomly divided into model group,CHYS low dosage group,CHYS medial dosage group,CHYS high dosage group and Fosinopril-treated group.The body weight,plasma glucose were measured.The liver pathomorphology was observed by HE staining and PAS staining, and type I and III collagen were measured with immunohistochemical technique.Liver GLUT-1 and TGF-β1 expression were mea- sured with reverse transeriptase-polymerase chain reaction (RT-PCR) and Western blot.Results:The glycogen accumulation and fibration were alleviated in the liver tissues of rats treated by CHYS,accompanying the sup- pression of type I and m collagen expression in the liver.Liver expression of GLUT-1 and TGF-β1 de- creased significantly compared with the model group(P〈0.05).Conclusions:CHYS have protective effect on ex- perimental liver injuries in diabetic nephropathy rats.The mechanism might be related to the suppression of expression of GLUT-1 and TGF-β1.
